dc.description.abstract | Biosurfactants are multifunctional compounds of microbial origin of industrial and
environmental interest due to their surfactant and emulsifying properties of biodegradable
nature and low toxicity. Several culture media and carbon sources are used in the production
of these compounds, as well as a wide variety of microorganisms, isolated from multiple
environments. The present work aims to promote a bioprospection in the microbiota
associated with fruit residues (orange, mango and fruit mix) in order to identify the genetic
composition of this microbiota related to the biosynthesis of biosurfactants. For this, the
residues were evaluated through a metagenomic approach, where the genetic potential of their
associated microbiota for biosurfactant biosynthesis was determined. The residues presented a
common core that includes the genera Klebsiella, Enterobacter, Stenotrophomonas,
Acinetobacter and Escherichia and similar functional profiles related to biosurfactant
biosynthesis, presenting a genetic potential mainly for lipopeptide biosynthesis, with
emphasis on genes related to the biosynthesis of putisolvins, mycosubitilin and iturin A. The
taxonomic assignments to the genes present in higher abundance in the samples were related
to the common core found. From the metagenome, 34 genomes (MAGs) were retrieved, 10 of
which were of high quality, which were evaluated for genes related to the production of
biosurfactants. MAGs showed genetic potential for the biosynthesis of biosurfactants,
especially lipopeptides, corroborating the metagenomic analysis. The MAG OB01
taxonomically identified as Brevibacillus laterosporus stood out for the number of assigned
genes. Subsequently, the microorganism isolated from mango residue, identified as
Brevibacillus borstelensis ISM04, was used in the fermentation process for the production of
biosurfactant using glycerol as a carbon source. The produced biosurfactant showed an
emulsifying activity of 67 % in kerosene and showed thermal stability up to 80 °C and at pH
range between 6 and 10, in addition, it did not show toxicity in tomato and lettuce seeds. The
compound produced was partially characterized and the analyzes suggest that it is a
lipopeptide biosurfactant. | eng |

dc.description.resumo | Biossurfactantes são compostos multifuncionais de origem microbiana de interesse industrial
e ambiental devido as suas propriedades tensoativas e emulsificantes de natureza
biodegradável e baixa toxicidade. Diferentes meios de cultivo e fontes de carbono são
utilizadas na produção desses compostos, bem como uma ampla diversidade de
microrganismos, isolados de múltiplos ambientes. Portanto, o presente trabalho teve por
objetivo promover uma bioprospecção na microbiota associada a resíduos de frutas (laranja,
manga e mix de frutas) visando identificar a composição genética da microbiota relacionada à
biossíntese de biossurfactantes. Para isso, os resíduos foram avaliados através de uma
abordagem metagenômica, onde determinou-se o potencial genético associado à biossíntese
de biossurfactantes. Os resíduos apresentaram um common core que inclui os gêneros
Klebsiella, Enterobacter, Stenotrophomonas, Acinetobacter e Escherichia e perfis funcionais
similares, relacionados à biossíntese de biossurfactantes, apresentando um potencial genético
principalmente para a biossíntese de lipopeptídeos, com destaque para genes relacionados à
biossíntese de putisolvinas, micosubitilina e iturina A. As atribuições taxonômicas aos genes
presentes em maior abundância nas amostras estavam relacionadas ao common core
encontrado. A análise metagenômica evidenciou que os resíduos de frutas podem ser
utilizados como fonte de microrganismos pois sua microbiota associada apresentou um
potencial genético para a biossíntese de biossurfactantes. A partir do metagenoma, foram
recuperados 34 genomas (MAGs), sendo 10 em alta qualidade, que foram avaliados quanto
aos genes relacionados à produção de biossurfactantes. Os MAGs apresentaram potencial
genético para a biossíntese de biossurfactantes, especialmente lipopeptídios, corroborando
com a análise metagenômica. O MAG OB01 identificado taxonomicamente como
Brevibacillus laterosporus destacou-se pela quantidade de genes atribuídos. Posteriormente, o
microrganismo isolado de resídio de manga, identificado como Brevibacillus borstelensis
ISM04 foi utilizado no processo fermentativo para a produção de biossurfactante utilizando
glicerol como fonte de carbono. O biossurfactante produzido apresentou atividade
emulsificante de 67 % em querosene e apresentou estabilidade térmica até 80 °C e entre pH 6
e 10, além disso, não apresentou toxicidade em sementes de tomate e alface. O composto
produzido foi parcialmente caracterizado e as análises sugerem ser um biossurfactante de
natureza lipopeptídica. | por |
