dc.description.abstract | Prochilodus nigricans is a migratory detritivorous fish, with a silvery grey colored body and
dark, irregular, wavy, bar-like patterns on the caudal-fin lobes. This species is widely
distributed in the Amazon and Tocantins river basins and stands out for its great economic
importance and social relevance due to subsistence fisheries. Because of the uncertainties
recently raised regarding the validity of the species, the aims of this work were to access the
genetic diversity of P. nigricans in the region of the Brazilian shield and to characterize the
genetic-population parameters of this species in the sub-basin of the Tapajós river during a nonreproductive period. In order to test the occurrence of hidden diversity, we used the
mitochondrial markers COI and ATP6/8 in samples from the Tapajós, Xingu and TocantinsAraguaia sub-basins, and conducted a GMYC analysis with COI, considering the sequences
generated in the present study and Genbank sequences of individuals sampled in other regions
of the Amazon basin. Population structure and genetic diversity analyzes were performed using
eight microsatellite loci and, the ATP6/8 genes and the mitochondrial DNA control region. For
both COI and ATP6/8, there was a significant divergence between individuals sampled in the
Tapajós river, with genetic distance values ranging from 1% to 1.9% between this group and
the other individuals sampled in the Brazilian shield. The GMYC analysis identified that these
individuals constitute another MOTU (molecular operational taxonomic unit), which were
excluded from the population analyzes conducted in the Tapajós sub-basin. Mitochondrial and
microsatellite data evidenced three genetically differentiated populations, Tapajós, Teles Pires
and Juruena, the latter of which was genetically more differentiated. The three evaluated
populations presented high and similar genetic diversity values to those of other species of
Prochilodus and showed no signs of population expansion or decline in the last 200 thousand
years. The results obtained in the present study on the diversity of P. nigricans are of particular
relevance, since they make possible a previous diagnosis of the genetic status of P. nigricans
populations in the tributaries and the main channel of the Tapajós river, before the eventual
effects caused by the implantation of dams. Such data will serve as a basis for future studies to
evaluate the impact of these constructions on the distribution pattern of the genetic diversity of
migratory species, serving as an important tool to observe changes in the genetic structure of
populations in areas fragmented by dams, and to subsidize management programs that aim at
the conservation of the ichthyofauna in these regions. | eng |

dc.description.resumo | Prochilodus nigricans é um peixe migrador, de hábito detritívoro e que apresenta corpo com
coloração cinza prateada, com faixas transversais escuras e inconspícuas na nadadeira caudal.
Esta espécie possui ampla distribuição nas bacias do rio Amazonas e do rio Tocantins, e
destaca-se pela grande importância econômica e social em virtude da pesca de subsistência.
Devido às incertezas recentemente levantadas acerca da validade da espécie, este trabalho se
propôs a acessar a diversidade genética de P. nigricans na região do escudo brasileiro e
caracterizar parâmetros genético-populacionais dessa espécie na sub-bacia do rio Tapajós
durante um período não reprodutivo. Para testar a ocorrência de diversidade oculta, utilizamos
os marcadores mitocondriais COI e ATP6/8 em amostras oriundas das sub-bacias dos rios
Tapajós, Xingu e Tocantins-Araguaia, e conduzimos uma análise de GMYC com o COI,
considerando as sequências geradas no presente estudo e sequências do Genbank de indivíduos
amostrados em outras regiões da bacia Amazônica. As análises de estrutura populacional e
diversidade genética foram realizadas para oito loci de microssatélites e para os genes ATP6/8
e a região controle do DNA mitocondrial. Tanto para o COI quanto para o ATP6/8 evidenciouse significativa divergência entre indivíduos amostrados no rio Tapajós, com valores de
distância variando entre 1% e 1,9% entre esse grupo e os demais indivíduos amostrados no
escudo brasileiro. A análise do GMYC identificou que esses indivíduos constituem outra
MOTU (unidade taxonômica operacional molecular), os quais foram excluídos das análises
populacionais conduzidas na sub-bacia do rio Tapajós. Os dados mitocondriais e de
microssatélites evidenciaram três populações geneticamente diferenciadas, Tapajós, Teles Pires
e Juruena, sendo esta última a que se apresentou geneticamente mais diferenciada. As três
populações avaliadas apresentaram valores de diversidade genética elevados e similares aos de
outras espécies de Prochilodus, não demostrando sinais de expansão ou declínio populaciona l
nos últimos 200 mil anos. Os resultados obtidos no presente estudo acerca da diversidade de P.
nigricans possuem especial relevância, uma vez que possibilitam um diagnóstico prévio do
status genético das populações de P. nigricans nos afluentes e calha principal do rio Tapajós,
antes dos eventuais efeitos ocasionados pela implantação de barragens. Tais dados servirão de
base para estudos futuros que visem avaliar o impacto dessas construções sobre o padrão de
distribuição da diversidade genética de espécies migradoras, servindo como um importante
instrumento para observar modificações na estrutura genética das populações em áreas
fragmentadas por barragens, e subsidiar programas de manejo que visem a conservação da
ictiofauna nestas regiões. | por |
