dc.description.abstract | Ciliates dominate microzooplankton and play an essential role in marine food
webs. By ingesting flagellates, bacteria and other organisms from pico and
nano-plankton, and then being consumed by zooplankton, ciliates are the main
pathway of carbon flow that links the microbial loop to the classical trophic
chain. However, the diversity and distribution of ciliates in the global ocean is
still poorly understood due to the difficulty of observing and classifying these
organisms under light microscopy. When fixed, most ciliates are lost or change
their morphology, making their identification difficult. Thus, the use of molecular
tools represents an opportunity to study the ciliate community in the ocean.
Within ciliates, the Strombidiidae family is amongst the most abundant, but
there are still few systematic studies at the global scale on the distribution,
abundance and phylogeny of this important group in the ocean. This study aims
to: 1) evaluate the phylogenetic diversity of Strombidiidae, 2) understand its
distribution in the global ocean, 3) determine the ecological niche of the most
abundant clades. We used a metabarcoding dataset (region V4 18S rRNA) of
1008 samples from the TARA Oceans and TARA Polar circum-navigation
expeditions. We constructed a Strombidiidae reference tree with complete 18S
rRNA gene sequences from the literature and inserted the metabarcoding
sequences for phylogenetic assignment. Then we related the distribution data of
these clades with environmental data to define ecological niches. The
phylogeny of Strombidiidae revealed 20 distinct clades, almost all of them
widely distributed in most oceanic regions. The most abundant clade
(Strombidium_clade2) is cosmopolitan. Furthermore, we obtained 12
environmental clades whose sequences (V4 18S rRNA region) are not identical
to those described in the reference tree, and thus, may represent new clades.
Niche analysis showed that the different clades of the Strombidiidae family had
different distribution patterns in the global ocean, determined by temperature,
salinity and oxygen. The Arctic Ocean had greater abundance among the
Strombidiidae clades and very characteristic niches were observed for these
oceanographic regions. This work represents the first study of the taxonomic
and phylogenetic diversity of the Strombidiidae family in the global ocean, thus
contributing to the knowledge of the ecology of marine ciliates. | eng |

dc.description.resumo | Os ciliados dominam o microzooplâncton e desempenham um papel essencial
nas teias tróficas marinhas. Ao ingerir flagelados, bactérias e outros
organismos do pico e nano-plâncton, e ao serem depois consumidos pelo
zooplâncton, os ciliados são o principal elo que liga a alça microbiana à cadeia
trófica clássica. No entanto, a diversidade e a distribuição de ciliados no
oceano global ainda é pouco conhecida devido à dificuldade de observar e
classificar estes organismos em microscopia ótica. Quando fixados, a maioria
dos ciliados são perdidos ou mudam a sua morfologia, dificultando a sua
identificação. Assim, o uso de ferramentas moleculares representa uma
oportunidade para o estudo da comunidade de ciliados no oceano. Dentro dos
ciliados, a família Strombidiidae é uma das mais abundantes, porém existem
ainda poucos estudos sistematizados na escala global sobre a distribuição,
abundância e filogenia deste grupo. Este estudo teve como objetivos: 1) avaliar
a diversidade filogenética de Strombidiidae, 2) entender a sua distribuição no
oceano global e 3) determinar o nicho ecológico dos clados mais abundantes.
Utilizamos um conjunto de dados de metabarcoding (região V4 18S rRNA) de
1008 amostras das expedições de circum-navegação TARA Oceans e TARA
Polar. Construímos uma árvore de referência de Strombidiidae com sequências
completas do gene 18S rRNA extraídos da literatura e inserimos as sequências
de metabarcoding para a designação filogenética. Depois relacionamos os
dados da distribuição desses clados com dados ambientais para a definição de
nichos ecológicos. A filogenia de Strombidiidae revelou 20 clados distintos,
quase todos amplamente distribuídos na maioria das regiões oceânicas. O
clado mais abundante (Strombidium_clade2) é cosmopolita. Além disso,
obtivemos 12 clados ambientais cujas sequências (região V4 18S rRNA) não
são idênticas às descritas na árvore de referência, e portanto representam
novos clados. A análise de nicho mostrou que os diferentes clados da família
Strombidiidae apresentaram diferentes padrões de distribuição no oceano
global, determinados pela temperatura, salinidade e oxigênio. O oceano Ártico
apresentou maior abundância entre os clados de Strombidiidae, com nichos
bem característicos para estas regiões oceanográficas. Este trabalho
representa o primeiro estudo da diversidade taxonômica e filogenética da
família Strombidiidae no oceano global, contribuindo assim, para o
conhecimento da ecologia dos ciliados marinhos. | por |
