dc.description.abstract | This study evaluated the influence of the degradation of a new hydrogel produced from EDTAD-modified starch in two different soils and the bacterial community involved in this process. The soils are an argisol, which was called "soil A", and a red oxisol, called "soil B". The soils were characterized and the argisol presented 12.4% of organic matter and pH 5.1, and the red oxisol, 11.6% of organic matter and pH 4.6. The argisol also has more residual soil moisture and higher amounts of micro and macronutrients, such as phosphorus, calcium, magnesium, and other physical characteristics such as cation exchange capacity, while the oxisol has more calcium phosphate and aluminum. The biodegradation test was performed using the Bartha respirometric method, measuring biodegradation by titrating the KOH solution that captured theCO2 emitted by the microbiota present in the respirometers with soil and hydrogel sample. It was observed that the biodegradation of the hydrogel in the argisol was more efficient, but more thorough and linear. After the end of the degradation experiments, the soil DNA was extracted from the DNA and sequenced for a qualitative analysis of the existing microorganisms. 340 to 480 genetic profiles were found in each one of the samples, with the highest diversity being in the soil sample B with hydrogel post-biodegradation, and the lowest, in soil A post-biodegradation. The fact that two different soils were used in this study shows how important their physicochemical properties are in the activity of the microorganisms present in the biodegradation process, since the sample incubated in the soil, richer in organic matter (albeit with a slight difference), micro and macronutrients and higher acidity obtained a more complete and uniform biodegradation, while soil B, with opposite characteristics, obtained a lower biodegradation of the hydrogel, even though it was incubated for a longer time, a fact that may have been influenced by a higher concentration of aluminium, which influences a lower bacterial activity. | por |

dc.description.resumo | Este trabalho teve por objetivo avaliar a influência da degradação de um novo hidrogel (produzido a partir de amido modificado com EDTAD) em dois diferentes solos e a comunidade bacteriana envolvida nesse processo. Os solos são um argissolo, o qual chamou-se “solo A”, e um latossolo vermelho, chamado de “solo B”. Ambos foram coletados em Araçoiaba da Serra, SP.Os solos foram caracterizados e o argissolo apresentou 12,4% de matéria orgânica e pH 5,1, e o latossolo vermelho, 11,6% de matéria orgânica e pH 4,6. O argissolo também apresenta mais umidade residual e quantidades maiores de micro e macronutrientes, como fósforo, cálcio, magnésio e outras características físico-químicas como capacidade de troca de cátions, enquanto o latossolo possui mais fosfato de cálcio e alumínio. O ensaio de biodegradação foi feito utilizando-se o método respirométrico de Bartha, aferindo-se a biodegradação através de titulação da solução de KOH que captou o CO2 emitido pela microbiota presente nos respirômetros com solo e amostra de hidrogel. Observou-se que a biodegração do hidrogel no argissolo foi mais eficiente, mas completa e linear. Após o fim dos experimentos de degradação o DNA do solo foi extraído do DNA e o sequenciamento para uma análise qualitativa dos microrganismos presentes. Foram encontrados de 340 a 480 perfis genéticos em cada uma das amostras, com a maior diversidade sendo na amostra de solo B pós-biodegradação, e a menor, no solo A pós-biodegradação. O fato de dois solos diferentes terem sido usados neste estudo, evidencia o quão importantes suas propriedades físico-químicas na atividade dos microrganismos presentes no processo de biodegradação, pois a amostra incubada no solo A, mais rico em matéria orgânica (ainda que com discreta diferença), micro e macronutrientes e maior acidez obteve uma biodegradação mais completa e uniforme, enquanto o solo B, com características opostas obteve uma biodegradação menor do hidrogel, mesmo tendo sido incubado por mais tempo, fato que pode ter sido influenciado por uma maior concentração de alumínio, que influencia para uma menor atividade bacteriana. | por |
