    This paper was prepared a material candidate for reference material for mineral mixture for dairy cattle. Shape certain analytes Ca, K, Na, P, Cu, Fe, Mn, Mg, Zn and contaminants As, Cd and Pb The material was crushed, sieved until a particle size less than 80μm and packaged. A study was conducted in the best sample preparation through the use of factorial design, planning Doehlert and 4 in a closed system open system experiments. Through a principal component analysis, we chose to sample preparation that was used in 3 mL of HNO3 acid 3mol.L-1 and 1 mL of H2O2 in a closed system. Techniques of absorption and emission spectrometry were evaluated for the determination of analytes in each of the study. Form testing for between-bottle homogeneity and within-bottle homogeneity to check if the sample is homogeneous. Were also carried out tests in the short-term and long-term stability. The values assigned to each analyte were obtained by an interlaboratory collaborative program. In the end, the uncertainties were determined for each stage of preparation of candidate material and obtained expanded uncertainty pair each of the analytes under study.

    Desenvolvimento de um sistema de produção recombinante de uma cistatina em uma linhagem industrial de Saccharomyces cerevisiae

    Saccharomyces cerevisiae is the most important microorganism used in alcoholic fermentation process. A strain of Saccharomyces cerevisiae widely used to produce alcohol in Brazil is the PE-2, due to its high fermentation capacity. The goal of this study was to develop an expression system for recombinant proteins using the industrial strain of Saccharomyces cerevisiae, PE-2. The protein chosen to be used as a model for this system was the Cane-CPI-1, an inhibitor of cysteine protease. Initially the construction of plasmids containing CaneCPI-1 gene was performed and yeast cells were transformed with pYADE4-CaneCPI-1 construction. The transformed strain was submitted to expression analyses during batch fermentative process with cell recycle. The recombinant protein was expressed in yeast cells and it was possible to purify the recombinant protein by affinity chromatography on nickel column. This purified protein was immunodetected using a polyclonal anti-CaneCPI-1antibody and monoclonal anti His-Tag antibody, which were also able to detect the CaneCPI-1 in a crude extract from Saccharomyces cerevisiae cells. Assays of inhibitory activity performed with the purified CaneCPI-1 revealed its ability to inhibit the catalytic activity of a cysteine proteinase. This study showed that the use of transformed strain did not affect the fermentation process and that the PE-2 industrial strain of S. cerevisiae can be a viable expression system for recombinant protein production and open perspectives for production of any protein of biotechnological applications during fermentation alcoholic process.

    Surface modification in implants is used, basically, to improve osseointegration, which is defined as the intimate contact between bone and implant material, without a soft tissue layer in the bone/implant interface. Titanium alloys, which are the most commonly used materials in the fabrication of hip stem, have an excellent biocompatibility. The surface modification of these materials can lead to a better osseointegration. The developments and studies of new modification processes are very dynamic and often related to biological aspects, for example, cellular growth on the modified surface. Studies regarding engineering and properties of the implant with modified surface are not up to date with the quickness of new processes developments and evaluation of these processes on cellular response. Since fatigue is a determinant property of implants, this study aims to evaluate the fatigue behavior of Ti-6Al-4V alloy with modified surfaces by laser and anodic oxidation. Thus, axial fatigue tests were performed to obtain SxN curves (Stress x number of cycles) of cylindrical specimens, in the polished and surface treated conditions. Roughness measurements and scanning electron microscopic (SEM) were used to characterize the features of the modified surface. As a result, the modification by anodic oxidation, for the parameters used, did not affect the fatigue behavior of Ti-6Al-4V alloy. Unlike anodic oxidation, the laser surface modification performed in this work decreases by about 30% the limit of fatigue strength of the alloy.
