    The objective of this dissertation is to study the narrative of the trailer of one of the most influential films in cinema history: Alfred Hitchcock´s Psycho (1960). The trailer will, therefore, be analyzed not only as a tool of movie marketing, but mostly as a 'shortmovie' which has a kind of parallel narrative and assist in the feature film, with a beginning, middle and end, trying to understand how that is constituted and how it still speaks to the disclosure´s posters and the opening credits of the film. Both 'core' sequence storyboard´s (trailer and movie) will also be used in this reading that seeks to comprehend the process of signification in the orbit of Psycho.

    The levels of Cu, Zn, Mn, Fe, Pb and Cd were determined in brown sugar from sugar cane produced by different crop systems in the organic and conventional systems. Pb e Cd were determined by atomic absorption spectrometry with electro thermal atomization in graphite furnace (GFAA), and the mineral metals Cu, Zn, Mn and Fe were determined by flame atomic absorption spectrometry (FAAS). Cu, Zn, Mn and Fe were detected but in levels below the limits recommended by the World Health Organization. The brown sugar from the conventional system without limestone but with chemical fertilizer showed the highest concentration of Cu, Zn, Mn and Fe, and that from the organic system with liming and organic compost showed the lowest concentrations. The levels of Pb found in samples from: conventional system with organic amendment and chemical fertilizer, and with liming and chemical fertilizer, and from organic system with liming and organic compost were higher than the tolerable limit for the Brazilian National Health Surveillance Agency for sugar. Cd was never detected. The results indicate the need of constant monitoring studies of the brown sugar produced in organic systems to assure its food safety.

    This master degree contains a literature review on the conservation of water, rainwater utilization, quality and water treatment, legislation and risk analysis. Was verified that rainwater utilization systems vary widely in type and would be important to be able to correlate the precipitated water, uses and models for each situation. And for this work was used the risk analysis focusing the preliminary hazard analysis. A bibliographic review carried out presented some guides and legislation that could complete quality control parameters for non-potable rainwater utilization, and determine the time to review these parameters to control the operation of the system. The study's preliminary analysis was used to study the greatest dangers in the use of rain-related water quality system, critical control points, the user risks, control measures and mitigation measures. For the case study was addressed the same preliminary hazard analysis for knowledge about the contamination of rainwater in relation to the control parameters and on their own knowledge of project development for rainwater utilization. Through the two analyzes a proposal of guidelines was performed for implantation of rainwater utilization system for the case study and there is a possibility of a real fitness for safe use.

    This work is aimed at investigating the inner life in Bergson s first book, Time and Free Will An essay on the immediate data of consciousness (1889). We try to analyze especially the notions of duration and space in light of what we call the divided self , i.e., the division established by Bergson between the superficial self related to space, social life and language and the deap-seated self which concerns the pure duration of consciousness, the continuous multiplicity of interpenetration of psychological states and creating and unpredictability that constitutes each free act. At the end, and already in possession of the conclusions on the Essay, we will discuss the bergsonian intuitive method and its relationship with the divided self.

    This work aims to discuss the representation of silence in films. Our objects of study are the films Cleópatra and A erva do rato both by Julio Bressane, since the phenomenon of silence emerges as a recurring theme in the style of this director. The analyzes found in this work focus on the discussion of the phenomenon linked to its discursive, and its physical appearance, and also about its manifestations in artistic fields like music, poetry and cinema. To do so, we had as main references studies of Paul Théberge, Fernando Morais da Costa, Virginia Flores about the sound in cinema, John Cage, Susan Sontag, José Miguel and Jorge Antunes Wisnik on the representativeness of silence for the arts in general.

    On the traversed course through the literature about institutional participation, it is possible to point distinct moments to deal with. In this dissertation was recovered the discussion approached by Carlos Estevam Martins, that in the end of the eighties and begin of nineties, bring forward the participation theme into the theory of the State and highlighted the relationship of participation and representation in this context. Following, we brought to the debate inspired research by the Habermasian theory, that pointed to an enthusiasm with the new forms of institutional participation. Subsenquently, research showed that was a large disparity between empiric bases and normative expectations in the literature until then. The last and current moment, focuses its efforts to the effective question of participation. During this period, many participation institutions (PIs) was analyzed and evaluated, however, the research about Public Hearings was rare. Based in the literature balance, this dissertation proposed to analyze Public Hearings, in the municipality of Araraquara-SP( Brazil), that was made throughout the revision process of the Master Plan of this city in 2013. The research point that the hearings, because of yours advisory character, worked as debate environments, but not as a political instrument of propositions and decisions. In addition, this Public Hearings operate as a subordinate participation instrument of government authority, because is that one who is able to incorporate or reject submitted suggestions by the population in this environment.

    The depletion of fossil fuels, the growing energy demand and the great need to reduce carbon emissions gradually increased the demand for new sources of renewable and clean energy. Bioethanol is obtained from the fermentation of lignocellulosic biomass, for example sugar cane, and can be considered a viable alternative. For this biomass can be used it is necessary to degrade the constituent molecules of the cell wall to fermentable sugars through the enzymes called cellulases. Among them there are three classes with different functions: the cellobiohydrolases, endoglucanases and β-glucosidases. In order to contribute to the viability and deployment of ethanol production technologies, purification of two β- glucosidases of the fungus Trichoderma harzianum was performed, named in this work as ThBgl1 e ThBgl2, as well as their biophysical and biochemical characterization. Clones in study were obtained by a cloning platform, expression and purification of recombinant proteins in high performance and expressed in bacteria Escherichia coli. Through the functional characterization of proteins under study it was found that they had the optimum pH in the range of 5.5 and optimum temperature around 40ºC. Results of enzyme kinetics showed that ThBgl1 has higher preference (specificity) by cellobiose, while in ThBgl2 cellobiose is a substrate having low specificity. In ThBgl2 specificity is higher by fucosideos. Furthermore, it was observed the occurrence of transglycosylation catalyzed by β-glucosidases in study and the action of some inhibitors on its enzymatic activity. The three-dimensional analysis of these proteins revealed the presence of the barrel tim typical of family 1 of glycoside hydrolases.

    The hypocretin or orexin A plays an important role in the modulation of respiratory control in mammals, but there are no data available for the role of Orexins in the peripheral and central chemoreception of non-mammalian vertebrates. Thus, the present study was designed to evaluate the location of orexinergic neurons in toads (Rhinella schneideri). In addition, we investigate if the orexinergic system of this species is important to hypoxic (5% O2 and N2 for balance) and hypercarbic (5% CO2, 21% O2 and N2 for balance) drive to breath. We assessed the role of the orexinergic system on respiratory responses by using intracerebroventricular injection of SB-334867 (orexin A receptor antagonist) during the light and the dark phase. Our results demonstrated that orexinergic neurons of Rhinella schneideri are located in the suprachiasmatic nucleus of diencephalon. Additionally, the injection of the orexin antagonist attenuated the ventilatory response to hypercarbia during the dark phase by acting on tidal volume and breathing frequency, while in the light phase, there was an attenuation in the ventilatory response to hypoxia by acting just in the tidal volume. We conclude that central orexin A contributes to hypercarbic and to hypoxic chemoreflex in toads Rhinella schneideri.

    O conceito de subjetividade em A Ideia da Fenomenologia

    The objective of this research is the study of the concept of subjectivity as such it develops in lessons on The Idea of Phenomenology (1907), by Edmund Husserl. Such concept in this work is linked to a critique of the traditional conception of representation, according to which the psychic immanence designate one intramental ontological region in which they find representations as substitutes for reference objects. To the extent that this criticism is made, the relationship between immanence and transcendence are redefined, providing a new phenomenological concept of subjectivity. This, in turn, is founded on the idea of authentic immanence, which includes not only sensations and acts (real [reell] immanence), but intentional objects in their modes of appearance (intentional immanence). This conception of subjectivity signals a change in relation to what had been advocated in the work initiating the Husserlian phenomenology - the Logical Investigations (1900-1). At this, subjectivity is presented as containing only the noetic character of knowledge - the real immanence -, while in the work studied here it also includes what subsequently one designate, in phenomenology, as noematic character of knowledge the immanence in intentional sense. The hypothesis of reading proposed here is that this conception of subjectivity is made possible by analysis of Husserl on time-consciousness, from 1905, in which it reveals that even within the "flow" of experiences, there is something identical, which beyond the scope of real immanence. This recognition would have led Husserl to the conception that the limitation of subjectivity to the sphere of real immanence is founded on a mistaken association between the latter and immediate evidence. Therefore, the route of Husserl's text in The Idea of Phenomenology will moves itself around the dissociation between these two notions. Draw will be, therefore, the path taken by Husserl in this work in order to highlight the notion of subjectivity that emerges in it.

    Musical knowledge, although usually conceived as instrumental expertise, has another important element: appreciation. Whereas most people can listen to musical stimuli, the way these stimuli are appreciated is largely influenced by individuals learning history. This study investigates how people with different learning histories concerning musical experience appreciate three performances of the same musical work recorded by performers of contrasting levels of expertise. To achieve this, three excerpts of a piano musical work were submitted to the appreciation of participants with discrepant levels of involvement with music (n=79). Participants task consisted of evaluate how much they liked each performance using a 7 point scale. The scores attributed to each performance were comparatively analyzed according to the educational level, listening habits and field of expertise of participants. Results point out that 1)the educational level of participants, as well as their field of expertise, correlates with the ability to discriminate between the performances presented and 2)in the case of non-amateur performances, the specific training of a musical instrument proved to be more decisive than other kinds of musical knowledge in order to help listeners to identify what is considered by musical community as a high level performance.

    Low and high LPS doses (endotoxin) have been used as models to study fever and endotoxic shock, respectively. In rats, systemic nitric oxide (NO) directly affects thermogenesis, but does not mediate those responses. Even though few studies in birds suggest NO as a pyrogenic signalling to brain, there is not enough evidence for confirming this suggestion. We hypothesise that, compared to mammals, systemic NO presents similar thermogenic action during endotoxin challenge in birds, even in early life. As precocial birds, chickens have a developed thermoregulatory capacity immediately after hatching, which constitutes an interesting model to study thermoregulation in early life. Thus, we investigated the role of NO in LPS-induced fever and endotoxic shock in 5-day-old chicks. The doses of 2 (LPS2) and 10 g kg-1 of LPS induced fever 3-5 hours after intramuscular (IM) injection, while 50 and 100 (LPS100) g kg-1 decreased Tc within the first hour followed by fever at 4-5 hours postinjection. Plasma nitrate levels increased 4 hours, but not 1 hour, after treatment with LPS2 and LPS100 (higher increase). There was no correlation between plasma nitrate concentrations and Tc at both LPS doses. L-NAME (non-selective NOS inhibitor; 50 mg kg-1; IM) inhibited LPS2- and LPS100-induced fever and increased the magnitude of the fall in Tc caused by LPS100. In contrast, no effect of L-NAME was observed in LPS-treated chicks in warm conditions. L-NAME decreased oxygen consumption in LPS2- and LPS100-treated chicks, but the response was more pronounced with LPS100. Moreover, LPS-induced huddling (heat conservation behaviour) was accentuated by LNAME. Our results seem to indicate that, like in rats, systemic NO is not a mediator of fever and endotoxic shock in chicks, but does affect thermogenesis in these animals, which may constitute a common effect of NO in the endothermic vertebr.

    Ajustes da variabilidade da frequência cardíaca em repouso e durante o exercício em indivíduos com asma controlada

    Asthma is a chronic disease, with periods of exacerbation and worsening of airflow obstruction and may be triggered by various stimuli, including exercise, and is associated with dysfunction of the autonomic nervous system (ANS). In this context, two studies were conducted to assess adjustments of ANS in asthmatic volunteers in different situations. The first study was entitled: Increased sympathetic modulation and decreased response of the heart rate variability response in controlled asthma , was designed to evaluate the adjustments of indices of autonomic modulation of heart rate (HR) at rest in supine and seated positions and during the respiratory sinus arrhythmia maneuver (M-RSA). The assessment of 12 asthmatic volunteers included pulmonary function tests and measures of heart rate variability (HRV). The results suggest that in asthma appears to be an association of airflow obstruction with reduced total variability, and in addition, to postural change seems to be reduced adjustment of the sympathetic nervous system in asthmatic women. The second study entitled: Airway responsiveness at exercise recovery and adjustments of heart rate variability at rest and exercise in controlled asthma , evaluated the adjustments of cardiac autonomic modulation during exercise its association with the exercise induced bronchoconstriction. Sixteen asthmatics volunteers underwent constant speed exercise test, forced vital capacity maneuver before and after exercise testing and register of HR and R-R intervals. With this second study, it can conclude that in asthmatics, even with controlled disease, injury presents postural and autonomic adjustments during the year and reduced complexity of dynamical systems of HRV during exercise.

    The international airports has been committed globally with the adoption and management of environmental sustainability practices, encouraged not only by the requirement contained in local laws, but also on the instructions of non-governmental organizations such as the United Nations (UN); by tax incentive policies; by positive results, proven and reported in case studies; and by instructions of international institutions dedicated to the certification of environmental sustainability in the built environment. In this way, this study sought to compile and present case studies relating to environmental sustainability actions taken at airports worldwide and present the main certifications of environmental sustainability for the built environment. After this overview, this work focused on research in the rational use of water, addressing the consumption of water issues in airports and identifying the environmental sustainability requirements, for that focus, recorded in the certification Leadership in Energy and Environmental Design (LEED) and Process AQUA-HQE - High Environmental Quality, in the database of the Sustainable Aviation Guidance Alliance (SAGA), and Memorial Sustainable Criteria for Enterprises (MCSE), prepared by Infraero as a basic guide for the development of sustainable projects of Brazilian airports under its management. To the identified requirements were relate actions, which were compared for the verification of similarities and common goals. This comparison gave a concise set of actions aiming the rational use of water resources, also by reducing the water consumption through the use of economizers sanitary equipment. This has enabled a suggestion to adapt the MCSE, by the incorporation of new actions, for example; as well as the application of some of these actions to sanitary uses at passenger terminal of Vitoria/ES Airport through the development and analysis of scenarios that have concluded the capability of sustainable actions in reducing the demand for water for sanitary purposes in passenger terminals of airports.

    The production of second-generation ethanol by enzymatic hydrolysis of biomass is considered a viable and promising alternative to face the global energy crisis and to decrease our dependence on fossil fuels. Therefore, it is necessary to degrade the constituent molecules of the plant cell wall such as lignin, cellulose and hemicellulose to fermentable sugars. However, the use of enzymes for this purpose is still expensive, leading to the increase on studies seeking to make them more feasible economically and technically. The present study aimed the molecular, structural and functional characterization of the endoglucanase Cel12A from the fungus Gloeophyllum trabeum by different techniques. Biochemical data revealed the substrate specificity for the enzyme and showed that β-glucan is the best substrate for its activity (239.2 ± 9.1 U mg-1). Optimal conditions for activity were pH 4.5 and temperature of 50 oC. Thermal stability assay indicated a half-life of 84.6 ± 3.6 hours at 50 oC. The kinetic parameters Km (3.2 ± 0.5 mg mL-1) and Vmax (0,40 ± 0,02 μmol min-1) were determined using β-glucan as substrate. Analysis of scanning electron microscopy of oat spelts and filter paper samples submitted to the hydrolysis by GtCel12A evidenced the degradation effects of these substrates compared to control samples. Moreover, the low-resolution envelope and the crystallographic structure for GtCel12A were determined. The structure revealed a β-sandwich fold with two β-sheets (A and B) and three α-helices, while sheet A showed five strands and sheet B nine strands. The comparative analysis of the amino acid sequence and homologous structures prompted us to identify the catalytic residues, Glu142 and Glu227 in the active site of the enzyme. These results are important for understanding and elucidating the enzyme molecular mechanism of action and other glycoside hydrolase family 12 endoglucanases.

    The shape memory effect represents a physical property of materials with great potential applications in several sectors of Industry. In metallic materials, it’s a phenomenon associated to martensitic transformation and its reversion process in which the material, after being deformed over its elastic limit, can reestablish its original form through heat treatment. It’s really worthwhile to explore this feature because it allows the fabrication of joints and pipe coupling joints without the use of welds. The stainless Fe-Mn-Si-Cr-Ni alloys which this property are potentially eligible for this application and thus have attracted attention of researchers because they’re a cheap economical alternative and easy to be produced when compared to copper and Nickel-Titanium alloys with shape memory shape effect. However, these stainless alloys still feature discrete values of shape recovery and limited corrosion resistance. Besides that, the fabrication processes of this alloys reported in literature involve the production through induction vacuum furnaces that add great cost to the final product and imply few engineering applications. In this regard, the present work aimed to contribute to the study of Fe-Mn-Si-Cr-Ni alloys with shape memory effect and creating and characterizing an alloy obtained by fusion in inductions furnace without controlled atmosphere, assessing the different conditions of material processing when in terms of microstructure, mechanical properties, degree of shape recovery and corrosion resistance. The fusion of the alloy has generated an ingot with a high level of non-metallic inclusions and a certain amount of interdendritic porosity as well as the formation of secondary phases in the microstructure. The homogenization treatment has dropped some of the secondary phases, but the air cooling generated precipitation in grain boundaries. In the heat-treated condition, the alloy featured better results in terms of shape recovery and corrosion resistance.

    Due to the strong influence of globalization, researchers and entrepreneurs have recognized the need and the importance of developing new approaches in order to increase the competitiveness advantage in new products development. In this increasingly competitive scenario, it is highlighted that the companies have to organize themselves to develop products in an efficient, fast and proper manner in the first time. The Failure Mode and Effects Analysis (FMEA) has to be conducted to determine potential risks and evaluate them in relation to its relevance, probability of occurrence and possibility of detection. These risks have to be minimized by introducing preventive actions, considering that the FMEA is an important tool for defects prevention in products. Therefore, the objective of this study is to present the introduction of a more efficient and effective method to perform the FMEA report, proposing improvement actions to speed up its preparation, thus reducing development time and eliminating deviation of interpretation, because for different cross-functional teams, different interpretations in the index of severity, occurrence and detection have been diagnosed for the same products and / or similar applications. As a result, this work adopted the methodology of action research. Based on this study it was suggested a FMEA standardized by process, where all manufacturing processes existing in the studied organization were identified and mapped. This standardization provided a significant time reduction to perform the FMEA from six to two meetings with the cross-functional team and also allowed quick, efficient and systemic feedback looping, consequently gaining productivity and reliability in the product development process as result.

    The studies presented herein show the use of molecular modelling coupled to spectroscopic data to tackle four different problems in organic chemistry. In the first study, the sulfonylation of the primary hydroxyl group of the compound (2R,3S,6S)-2-(hydroxymethyl)-6-(1-phenyl-1H-1,2,3-triazol-4-yl)-3,6-dihydro-2Hpyran- 3-ol was addressed. The results show that the occurrence of the reaction depends on the stability of the S Cl secondary interactions and of the C-H Cl hydrogen bonds along the reaction pathway. The second investigation was the cycloaddition between benzyl azide and (organotelanil) alkynes. This reaction has a high driving force towards the products and generates fast and selectively a triazole with a n-butyltelluride substituent attached at position 5, which is stabilized by CH...Te interactions. So this cycloaddition fits the requirements of Click Chemistry. In third place, the conformations and relative configurations of 7 and 9 membered rings were elucidated by evaluating steric strain. The 7 membered ring shows a twisted chair conformation (TC) with C1, C7 and C10 having relative configurations S, R and S. The 9 membered ring has a twisted chair-chair conformation (TCC) and relative configurations R, S of C7 and C10. The fourth study was the optimization of rotational conformations of 1- (arylsulfonyl)indole derivatives. The different optimized conformations have the same stability and low rotational energy barrier, so that in the solid state the number of independent molecules in the asymmetric unit essentially depends on the packing intermolecular interactions. Regarding the stability of sulfonamide bonds the importance of hyperconjugative effects were showed, and the origin of the rotational barrier in the S-N bond is explained by the difference in steric hindrance and electrostatic interactions in the transition and the fundamental states.
