    The tahiti lemon belongs to the family Rutaceae, Citrus of the genus, species Citrus Latifolia, the variety Tanaka. It is a heterogeneous fruit, formed by three distinct fractions, them being the epicarp (outer shell), the mesocarp (white layer), and endocarp (pulp with buds and vesicles containing its juice). Both the whole lemon as its fractions have several culinary applications, and also in the food and pharmaceutical industry. Therefore dehydration of this product for food has as challenges, preserve good visual appearance and the components of interest at the end of the process. The objective of this study is to analyze the drying of each lemon fractions tahiti (epicarp, mesocarp and pulp), alone, and also of the slices at different temperatures and to evaluate color changes and shrinking occurring in the drying. Furthermore, verify the fits for the drying kinetics curves to diffusive models, empirical and semi-empirical and also test the use of ANNs for adjustment and simulation of drying. The drying tests were conducted in a natural convection oven with air at temperatures of 30°C, 40°C, 50°C, 60°C and 70°C. It was found that the epicarp and mesocarp fractions were quickly reached dynamic equilibrium moisture, followed by the endocarp, and the slices showed shorts rates of drying. Under the conditions evaluated for endocarp sliced, and the slices at 70°C there was a reduction drying time of about 90% over the time required at 30°C, and the fractions that make up the shell this reduction was approximately 66%. The fits of the drying curve to the diffusive kinetic model were reasonable. Adjustments data to empirical models in general were good, and among the tested models, models of Page and Overhults presented the best fits the drying kinetics. The ANNs showed a promising tool in the prediction and simulation of drying kinetics, but a larger number of data is needed to improve their performance. The color change occurred increasingly with increasing drying temperature, being higher for the endocarp and the slice. The shrinkage was higher in the early stages of drying, and reducing the surface area of the samples was about 33%.

    The increase in the municipal and industrial waste generation has caused enviromental and public health problems and as a consequence laws exist to address the issue. In Brazil, with the Federal Law No. 12,350 / 10 about the Política Nacional de Resíduos Sólidos reverse logistics (RL) of some products has become mandatory, including waste tires. However, RL of waste tires has been structured since environmental government agency resolutions propose treatment for tires disposal incorrectly in the environment and new generation of waste tires. Currently, the reverse network is managed by the tire manufacturers and importers associations, and destinations are commonly used are co-processing in cement kilns, and the recovery of rubber and steel as secondary products, by processes such as scrapping or lamination. Increasing transportation and operation costs in the logistics network make the network design critical for the full compliance with the legal goal. This network design is aligned with a planning that considers the financial issues like the minimization of costs or the maximization of profit, while meets the requirements of environmental government agencies. So it involves key strategic decisions, as the location of facilities and material flows, taking into account many parameters simultaneously. The objective of this study is to propose a model for waste tires reverse logistics considering: (i) flows from the output of the collection points (called ecopontos ) to the destination companies; (ii) the possibility of processes as the sorting of used tires in usable condition or not; (iii) the grinding process as an intermediate phase and for which type of destination the tire would be sent. Still are considered fixed costs for the installation of storage centers, for sorting used tires, and intermediary companies, for grinding and separation of components, in addition to the variable operating costs, transportation and potential revenues generated from the substitution of raw materials or fuel in destination companies. The model is a mixed integer linear problem (MILP) with multiple time periods. Experiments are done with a single and multiple periods, finally were present some sensitivity analysis. Other financial constraints as the annual budget and an approach that includes the carbon footprint (CO2) in the transport and processing are explored. The results showed configurations that meet the goal and have a small profit, indicating that storage centers are preferred in places far from destination points and low demand, while the pre-processing companies have opposite behavior. When using the carbon footprint, it was found that the sorting of used tires gain more importance in the view of reducing emissions, because there is less emission in the reuse or refurbishment compared to the manufacturing of new tires.

Thus, this work aims to analyze the poetic work in Dinâmica Subtil of António Ramos Rosa, through the Theory of Landscape, demonstrating how the literary landscape shows itself in the work and through witch resources it opens space for the construction of the literary landscape by the reader; this way, it aims to demonstrate how the literary landscape given in Dinâmica Subtil is formed and how can it be transformed by the reader.

    Currently the great challenge for the production of second generation ethanol is to reduce thecost of the enzymes. It is possible to reduce part of this cost by carrying out an optimization ofthe process fermentation using sources of cellulase-induction that allow further growth of fungalbiomass, and increased secretion of proteins. The use of a mutant strain with overexpression ofhemicelulolytics activators could increase expression of the enzymes of interest and therebycontribute to cost reduction. This work aimed to study the production of enzymes involved inthe degradation of biomass by the newly isolated strain of Trichoderma harzianum P49P11focusing on the improvement of the submerged fermentation processes and on the use molecularbiology tools to improve the fungal strain. Regarding the fermentation process, the effects ofdifferent inducing sources were evaluated in flasks and bioreactor using statistical experimentaldesign tools and strategies to enhance biomass in the pre-culture step. For fungal strainimprovement, it was used molecular biology tools for the overexpression of two activators ofcellulases (xyr1 and lae1). A proteomic analysis of the T. harzianum enzymatic extract obtainedusing sugarcane bagasse pretreated by steam explosion followed by delignification (BED) wasperformed. The results showed that the best source for inducing cellulase was BED + sucrose(3: 1), reaching values of 1.21 FPU/mL 80.0U/mL of xylanase and 17.30 U/mL of β-glucosidase. The proteomic analyis identificated 24 different glycoside hydrolases and fourCBM proteins, within 12 different CAZy families. From this study, the enzymatic cocktailproduced "on site" could be supplemented using two accessory enzymes, pectinase and α-Larabinofuranosidase,leading to an increase of 100% of the hydrolysis yield. Regarding thestudy of carbon sources in the pre-culture step, it was possible to increase cellulases productionin 2 times using glycerol as the initial carbon source, followed by inducing carbon source(BED). The xyr1 and lae1 overexpression influencied positively the FPase, CMCase, xylanaseand β-glucosidase production, representing a new approach to increase production of theseenzymes.

    The continuous demand growth for liquid fuels, alongside with the decrease of fossil oil reserves, unavoidable in the long term, induces investigations for new energy sources. A possible alternative is the use of bioethanol, produced by renewable resources such as sugarcane bagasse. Two thirds of the cultivated sugarcane biomass are sugarcane bagasse and leaves, not fermentable when the current, first-generation (1G) process is used. A great interest has been given to techniques capable of utilizing the carbohydrates from this material. Among them, production of second generation (2G) ethanol is a possible alternative. 2G ethanol requires two additional operations: a pretreatment and a hydrolysis stage. Regarding the hydrolysis, the dominant technical solution has been based on the use of enzymatic complexes to hydrolyze the lignocellulosic substrate. To ensure the feasibility of the process, a high final concentration of glucose after the enzymatic hydrolysis is desirable. To achieve this objective, a high solid consistency in the reactor is necessary. However, a high load of solids generates a series of operational difficulties within the reactor. This is a crucial bottleneck of the 2G process. A possible solution is using a fed-batch process, with feeding profiles of enzymes and substrate that enhance in the process yield and productivity. The main objective of this work was to implement and test a system to infer online concentrations of fermentable carbohydrates in the reactive system, and to optimize the feeding strategy of substrate and/or enzymatic complex, according to a model-based control strategy. Batch and fed-batch experiments were conducted in order to test the adherence of four simplified kinetic models. The model with best adherence to the experimental data (a modified Michaelis-Mentem model with inhibition by the product) was used to train an Artificial Neural Network (ANN) as a softsensor to predict glucose concentrations. Further, this ANN may be used in a closedloop control strategy. A feeding profile optimizer was implemented, based on the optimal control approach. The ANN was capable of inferring the product concentration from the available data with good adherence (Determination Coefficient of 0.972). The optimization algorithm generated profiles that increased a process performance index while maintaining operational levels within the reactor, reaching glucose concentrations close to those utilized in current first generation technology a (ranging between 156.0 g.L⁻¹ and 168.3 g.L⁻¹). However rough estimates for scaling up the reactor to industrial dimensions indicate that this conventional reactor design must be replaced by a two-stage reactor, to minimize the volume of liquid to be stirred.

    The pain is an universal experience, and frequently follows anxiety and depression cases. Some specific drugs used in depression and anxiety treatment are also used for the relief of pain. There are evidences showing that exposure to threatening situations can result in pain inhibition. In this way, studies involving pain and anxiety use animal models as tools for potential therapeutic agents screening as well as for enhance the knowledge about the neurobiology of emotions. This study investigated the following effects: acute and chronic fluoxetine treatment; microinjections of 5-HT1A and 5HT2C receptors agonists intraperiaqueductal gray matter (PAG), and the administration combined of these drugs on antinociception induced by the open arms (OAA) of the elevated plus maze (EPM). Furthermore, we investigated the effects of fluoxetine chronic treatment on serotonin, 5- HIAA, 5-HT1A and 5HT2C receptors levels within PAG. Our results showed that: (I) fluoxetine acute treatment (20 mg/kg) was able to increase the OAA; (II) fluoxetine chronic treatment (21 days) (20 mg/kg) also enhances the OAA, and decreased the number of writhes on animal confined in the enclosed arm, featuring an analgesic effect; (III) the administration of 8-OH-DPAT (5-HT1A agonist) intra-PAG did not change the number of writhes in animals confined in the arms of the EPM; (IV) the infusions of mCPP and MK- 212 (5HT2C agonists) intra-PAG increased the OAA. (V) The blockade of 5HT2C receptors located in the PAG by SB 242080 injection (5HT2C antagonist) was able to completely reverse the OAA; (VI) the combined administration of fluoxetine (acute treatment) and 8- OH-DPAT intra-PAG also blocked completely the OAA; (VII) the combined administration of fluoxetine (acute treatment) and MK-212 intra-PAG reversed the increase of OAA observed with MK-212 injection; (VII) combined administration of fluoxetine (chronic treatment) and 8-OH-DPAT intra-SCP once more blocked OAA; (IX) the fluoxetine (chronic treatment) + MK-212 intra-PAG, impairs the OAA increase observed with isolated MK-212 injection; (X) fluoxetine chronic treatment was not capable to change the serotonin, 13 5-HIAA levels, and serotonin turnover in the PAG (XI) fluoxetine chronic treatment promoted up-regulation of 5-HT1A and 5HT2C receptors within PAG. These findings suggest that OAA is médiated by 5-HT2C receptors located in the PAG. Furthermore, fluoxetine was capable to interact with this serotonergic receptor promoting functional (acute treatment) and quantitative (chronic treatment) changes on 5-HT1A and 5HT2C receptors within PAG.

    Social behavior can be defined as the behavior of two or more people, one over the other, or all together through a common environment. The aim of this study was to identify if the pictures that represent plays and games could function as reinforcers to typical children and also for children with deficits in social behavior and learning problems. In the first experiment, the participants were six children with deficits in social behavior and learning problems. The experimental stimuli were two geometrical figures and 12 pictures of two adults playing. Daily preference assessments were made, and then, the participants performed a task in which they had to chose between two geometrical figures, a square or a circle, with one of the figures assigning with a S+ function while the other with a S- function. After the participants select one of the two geometrical figures they could have the opportunity to engage in play represented picture considered the most preferred on the previous preference assessment phase. In the second experiment, the participants were three children with typical development and the experimental stimuli were the same geometrical figures used in the first experiment, and new pictures that representing games (apps) for Tablet. Following the preference assessment, after selecting a geometrical figure, was given to the participants the opportunity to engage in the game with the Experimenter 2, represented in the most preferred picture of them. The dependent variable was the percentage of times that the participant chose a particular geometrical figure per session. The results of this study demonstrate that play and games as a reinforcing function can be identified using a preference assessment with pictures. This procedure can be conducted quickly and with relatively low cost response. This research demonstrated the importance in of offering opportunities for choice and contact with plays for typical children and also children with deficit in social behavior and learning problems shown that plays and games were reinforcers.

    Memória discursiva da ditadura no século XXI: visibilidades e opacidades democráticas

    In this study, we start from the observation that, in distinct positions, different memories about the dictatorial times are brought back to present times. On the one hand, the slow unfoldment of official documents from the Brazilian dictatorial times and the refusal to revise the Amnesty Law – established in 1979, during the process of political opening initiated by the regime – and, on the other hand, a large amount of material produced by the media about the period or related to its themes build a discursive confrontation that introduces the production (by the media) of a memory inserted between the democratic (the possibility of “saying everything”, and the debate on the official level) and the silence (saying it on the margins). Yet, the revision process of the Amnesty Law, in 2010, and the implementation of the National Truth Commission, by the Federal Government, in 2012, serve as the base events to create the possibility of commenting, analyzing and rewriting history, in addition to the production of this/these memory/memories. Based on the theoretical propositions of the French approach of Discourse Analysis, in line with studies on the memory of different fields of the human and social sciences, we analyze in this study three types of materials: a) texts that were produced in the dictatorial period, especially by the independent media, that are constantly reproduced, republished and currently published (such as the magazine Pif Paf and the newspapers O Pasquim and Ex-); b) special editions of the magazines and newspapers published in the XXI century, besides documentaries and films that deal with facts relating to that period; c) texts produced by the media resulting from the revision process of the Amnesty Law and the establishment of the National Truth Commission. With that, we seek to understand the role of memory to the production and (re)construction of the history of the period in which Brazil was under military dictatorship, and the deepening of the democratic society. As a result, we understand that memories of the Brazilian military dictatorship are formed from a reordering, between opacity and visibility, of the ways of saying; in these 30 post-dictatorship years new discursive regimes emerged, enabling „to say the dictatorship‟, which sets, in Brazil, a will of memory and a step towards the consolidation of its democracy.

    Breast cancer is the leading cause of death in Brazil and worldwide, between the types of cancer, triple negative TNBC (Triple Negative Breast Cancer), which do not express hormone receptors, represents 20% of the cases and presents relapses within 3 years. However, the major cause of death in cancer is due the formation of metastasis. The TNBC has a greater propensity to form lung and brain metastasis. Furthermore, currently few treatments are available. Search for new target treatments, with fewer side effects is very important to treat this disease. Natural products are rich sources of molecules with antitumoral activity; among them are the gingerols, from ginger and resveratrol, from grapes. Nevertheless, not much is known about antitumor activity of [10]-gingerol (10G). Therefore, the aim of this work was to investigate the potential use of 10G as an antimetastatic molecule in in vitro and in vivo experiments. In this work, 10G had antiproliferative activity on several breast cancer cell lines (4T1BM2, 4T1BM2 shRNAβ3, 4T1Br4, MDA-MB-231, MDA-MB-231 BrM) and on a normal cell line, bEnd.3. The natural compounds affected tumor cell morphology and RSVT was able to inhibit cell migration and adhesion through vitronectin. 10G induced apoptosis via the extrinsic pathway through the increase of caspase-9, -3 and -7 expression and decrease of Bcl-2 expression, induced cell cycle arrest in G1 and subG0 phase. In in vivo models, 10G inhibited primary tumor growth and lung metastasis, as well as bone and brain metastasis. Therefore, this study was able to provide new data as 10G can be acting in tumor cells and its possible clinical application.

    In general, the idea of this work is to investigate the paradox, our object, by a linguistic and argumentative way, from the Theory of Semantic blocks (TBS). And specifically, for this, we adopted as main methods, two gestures: the relationship between linguistic elements as analysis condition, on the one hand, and the interdependence of its structure and significance, on the other hand, both in the non- use (language) and the use of language (enunciation). This second method of interdependence, when applied to our object, the paradox, becomes our first central hypothesis, the possibility of semantic paradox: the strange and difficult linguisticargumentative consideration of opposite significances, but in the interdependent state. And in this context we will also develop a second central hypothesis: the argumentative cube, while theoretical actualization of the argumentative square (the basic theoretical framework of our theory). Organizationally, the resourcefulness of this work will be carrying out in several specific strands, distributed throughout the chapters, namely: theory / paradox (divided into "Argumentation in the Language" / paradox, at first, and "Theory of Semantic Blocks "/ paradox, in a second stage); discours / paradox; dictionaries / paradox; "Historic Semantics of Enunciation" / paradox; relationship between languages / paradox and anteriority / paradox. In addition to these relational axes established to better observe our object, the paradox, and defend our hypothesis, the semantic paradox, we elected as central corpora, a group of dictionaries in which we can catch "which is said to be" a paradox and "as they say "a paradox to ponder" which means "a paradox. These dictionaries are divided into main corpus: Dictionnaire Historique de langue francaise, and and secondary corpora : Enciclopédia e Dicionário ilustrado, Kokugo Ziten e Dicionário Houaiss da Língua Portuguesa, among others.

    This thesis aligns with the idea that language teaching and learning is intrinsically linked to cultural issues, and that both the student and the teacher should think about this relationship during an intercultural learning process. Culture is a question of otherness: putting yourself in someone else’s place and, at the same time, assuming yourself as individual, as part of a different socio-cultural context in an exercise of translation and self-translation. Thus, we believe that the translation of songs and literary texts in English learning and teaching, for instance, could offer students the possibility of thinking, during the act of translation, about the lexicon and structures of the foreign language and the mother tongue, contributing to the enhancement of the training process and introducing them to creative work, as well as enabling contact with cultural issues of English-speaking countries from the study of lyrics and literature, rich sources of information on socio-cultural and historical topics. Therefore, at first the researcher aimed at investigating and discussing the representations (MOSCOVICI, 1997, 2013; JODELET, 1997) that students and teachers (in training and in service) carry on the act of translation, presenting exercises and courses in order to promote theoretical and practical awareness on the subject. After this preliminary study, in-service teachers who participated in one of the courses were chosen as subjects for further research and data analysis on the role of literary translation exercises in classroom in order to promote interdisciplinary reflection on the foreign language/culture and, at the same time, on the student’s own maternal language/culture following the concept that translating can mean interpreting, and that the translator plays a crucial role of second author who rescues meaning and transports them to the other side, connecting the two plans in a cultural otherness exercise (PAZ, 1976, 1990; DERRIDA, 1998; CAMPOS, 1992, 2013a, 2013b, 2013c; ARROJO, 1992, 1993, 2002; OTTONI, 1997, 1998; etc.). Finally, in general, in addition to the commented objectives, the intention was to contribute to the spread of the reading practices of various genres, to the appreciation of translation and the translator, to the reflective learning process of in-service teachers and for further expansion of the area of foreign and first language teaching and learning, seeking the best future for the area in Brazil.

    The benefits and the costs of the monetary policy alternatives have been debated trough decades, without a consensus of how it should be conducted. The most common instruments are, between other, the interest rate, the money supply and monetary reserves. In the last twenty five years, the generalized adoption of inflation targeting evidenced that monetary policy had been based only on the interest rates, and the new regime demanded measurement on inflationary expectations and inflation sensibility on interest rate changes. However, the monetary aggregates observed signs brought back the discussion of its use as an instrument, since the European Central Bank (ECB) had put weight on the aggregates behavior to its monetary policy decisions. Besides it, Brazilian monetary policy neglects the usage of the monetary aggregates as suggested by the ECB. Therefore, this thesis aims to verify the effects of monetary shocks from Divisia and simple sum monetary aggregates on Brazilian product and inflation considering the period between February 1980 and December 2013. Each asset that compose the monetary aggregates were weighted by its opportunity cost obtaining the Divisia monetary aggregates indexes. Vector auto regression models (VAR) were estimated to study the relationship among variables. Specially, Divisia monetary aggregates showed higher sensibility in each economic scenario considered, even with similar behavior to the simple sum intensities. This seems to be intensely observed on the Divisia monetary aggregates impulse response functions. So, considering the weighted monetary aggregates, the impacts on the product and price level seems to be sensible to the economic scenario according to each economic scenario particularity.

    The gull Larus dominicanus is a common shorebird in south and southeast of Brazil. Being opportunistic regarding the occupation of habitats and type of food, can expand their populations. After the reproductive period occupies beaches, estuaries and mangroves, where they feed and spend part of their life cycle. Aspects involving the reproductive biology of this species have been studied in this state, requiring update for the proper management. However, there are still gaps on exposure to chemical and microbiological contaminants, and how these variables could influence the populations of the species. The objective of this thesis was to analyze the association of L. dominicanus to the pathogenic bacteria, their vulnerability to heavy metals and aspects of reproductive biology in three breeding sites, located on the coast of Santa Catarina. In the first chapter cloacal samples of young gulls were analyzed, indicating the presence of harmful microrganisms. In this article, we discuss the diversity of bacteria isolated from samples obtained in the islands, as well as its relationship with the feeding habit of this birds. The second chapter is about the analysis of heavy metal contamination in young gulls feathers. The results showed contamination by lead in two of the three islands studied. There is evidence that L. dominicanus is in process of bioaccumulation of metals, arising from the exploration of coal, developed near to the study areas. The third chapter compares reproductive aspects of gulls, as hatching success, posture, volume of eggs and offspring development. No differences were observed when comparing the reproduction between the islands, and the data were similar to those reported by other authors. Considering the three articles, it appears that L. dominicanus acts as a reservoir of pathogenic bacteria, and is vulnerable to contamination by heavy metals of anthropogenic origin, and no evidence of change in reproductive success and population dynamics of species.
