Item type:Item, Efeitos do treinamento resistido e da reposição de 17beta-estradiol em ratas ovariectomizadas na modulação de p-AKT e PARP-1 clivada no hipocampo(Universidade Federal de São Carlos, 2016-09-08) ;Lagoeiro, Cristiani Gomes ;Perez, Sergio Eduardo de Andrade ;https://lattes.cnpq.br/9408953831211309https://lattes.cnpq.br/2716213108878282Menopause is a physiological process associated with physiological changes that lead to increased incidence of some diseases such as sarcopenia, cardiovascular disease, diabetes mellitus and neurodegenerative diseases. Apoptotic process is essential for the maintenance and development of tissues. However, exacerbation of this process can lead increase to neuronal death in neurodegenerative diseases. Objective: To investigate the effects of resistance training and 17β-estradiol replacement on the modulation of p-AKT and cleaved PARP-1, proteins envolved in apoptotic process, in the hippocampus of ovariectomized rats. Methods: 66 female rats Holtzman were randomly divided into six groups: 1) Sham operated Sedentary (Sham-Sed); 2) Sham operated Resistance Training (Sham-RT); 3) Ovariectomized Sedentary (Ovx-Sed); 4) Ovariectomized Resistance Training (Ovx-RT); 5) Ovariectomized Sedentary with 17β-estradiol Replacement (Ovx-Sed-ER); 6) Ovariectomized Resistance Training with 17β-estradiol Replacement (Ovx-RT-ER). The RT protocol required the animals to climb a 1.1 m-long vertical ladder with weights attached to their tails. Each session consisted of 4 to 9 climbs with 2 minutes interval between climbing, performed once every three days for 12 weeks. Protein content of p-AKT and cleaved PARP-1 were analyzed by Western Blotting and concentrations of 17β-estradiol were determined by ELISA. Results: Our results indicate that Ovx-RT groups and Ovx-RT-ER had lower protein content of cleaved PARP-1 compared to Ovx-Sed groups and Ovx-Sed-ER respectively and higher protein content of p-AKT compared to the group Ovx-Sed in the hippocampus. Conclusion: The results of this study show that resistance training performed for 12 weeks, with or without ER may be an effective intervention to promote greater protein content of p-AKT associated with the pathway of cell survival and lower protein content PARP-1 cleaved associated with apoptosis pathway.Item type:Item, Estudo das propriedades ópticas de filmes finos e poços quânticos de GaAsPN/GaPN(Universidade Federal de São Carlos, 2016-09-08) ;Covre, Felipe Soares ;Galeti, Helder Vinicius Avanço ;https://lattes.cnpq.br/3876752605313288https://lattes.cnpq.br/0605656438457962Diluted nitride III-V semiconductor leagues have physical properties that make them interesting for applications on optoelectronic devices. The possibility to lattice matching GaAsPN with silicon makes this semiconductor interesting for studies. In this dissertation we investigated the optical and magneto optical properties of semiconductors nanostructures of the type GaP(N)/GaAsPN. Mesures of photoluminescence (PL), photoluminescence excitation (PLE) and magneto photoluminescence (Magneto-PL) under high fields (B≤ 15T) were performed in films of GaAsPN and multiple quantum wells (MQW) of GaAsPN/GaPN. We have studied localizations effects with measures of diamagnetic shift, stoke shift and the dependence of photoluminescence peak with temperature. We observed a negative diamagnetic shift for some samples, which is an anomalous effect in these systems. It was also seen a red shift of the PL peak when the MQW samples suffered a thermal treatment. As the monomer composition of the EPS is presented as heteropolymers often composed of a diversity of monomers and that the diversity of EPS between algae is huge and taxonomically close organisms each other have compounds slightly different, made us believe in their use for chemotaxonomy of microalgae. The use of EPS for this purpose, was due to the ease of extracting them from cultures, their facility to be analyzed, together with their specificity and their constitution by only one or two fractions. This study goal was to verify the possibility of using EPS as features for a possible chemotaxonomic approach. Thus the characterization of algae polysaccharides was essential for a better understanding of the differences and similarities found in the EPS. Thus, throughout the chapters we will look at the characterization of specific groups which had most representatives for the selected genres. These genera were: 1) Ankistrodesmus; 2) complex Selenastrum / Messastrum / Curvastrum; 3) Monoraphidium; and 4) Kirchneriella. The lack of bibliographic material thinning policy has been an obstacle for the administration of the library with the aim to remove the collection that is not interest of the users of the BCo and consequently make the better use of the library's physical space, such as the inclusion of new acquisitions and/ or expand study spaces to users. The aim of this mastering dissertation is to evaluate the bibliographic collection of BCo and propose a thinning policy to contribute to the rationalization of physical space. For this, are considered the lack of relevant circulation of the collection of the last 3, 7 and 10 years respectively, by existing knowledge area on the campus of UFSCar in San Carlos city. To achieve low porosity, which ensures the high performance of the product, is required the formation of large amount of liquid phases during sintering and the flow of these liquids are responsible for the undesirable pyroplastic deformation. Some factors exacerbate the problem: large and rectangular shapes and faster firing cycles. Moreover, the production of tiles through a more sustainable route, the dry route, lacks studies evaluating the effects of processing method on pyroplastic deformation. Thus, this study aimed to contribute to reducing the incidence of pyroplastic deformations in porcelain stoneware tiles, in order to identify the main variables involved and suggest maneuvers for achieve this goal. In the first stage, technical porcelain stonewares were formulated using common raw materials in this type of manufacturing and prepared by wet route. The main difference between the batches was that 10% of the feldspar was replaced with other raw materials such as potash feldspar and quartz in different granulometries in order to change the characteristics of the liquid phases and to minimize pyroplastic deformation. The characterization of the batches before and after firing, as well the assessment of physical and chemical / mineralogical effects of the raw materials on deformation behavior was essential to identify the variables prevalent in deformation. In second stage some batches from previous step were prepared by dry route method, and then the same tests were performed in order to evaluate the effects of processing route on pyroplastic deformation. The results indicated that batches prepared by dry processing route are less likely to deformation mainly due to heterogeneity of microstructure. An aspect in the use of panels in buildings, barely explored in Brazil, is the possibility of considering it actively contributing in responsiveness to loads imposed, interacting with other elements such as pillars, slabs and beams. Since it is known that the literature about the theme is very limited, and facing a new specific standard coming soon, ABNT NBR 16475 (2016), this study has as objective to review the concepts of design and dimensioning of precast wall panels. With a review of the most recognized literatures and previous research already made, as well as analysis of the current standards, is shown the major advances related to the research theme, as well the standards parameters and criteria used nowadays. Glasses are commonly known to be electrical insulators, but some of them may present high conductivity and be candidates for different applications. The more conductive glasses result from the dissolution of halogenated salts in the glassy matrix, causing its ionic conductivity to increase by several orders of magnitude. Our approach proposes that glass can be compared to a solution in which a dissolved halogenated salt (solute) is weakly dissociated in the glassy matrix (solvent). This approach, called the weak electrolyte model, was initially proposed in the 70s to explain the almost exponential increase in the ionic conductivity of glasses in response to increasing concentrations of network modifiers (alkaline oxides). Our work proposes to expand this approach, correlating the increase in ionic conductivity with the increase in the thermodynamic activity of AgI. In addition, experiments were carried out at different temperatures in various glass compositions to confirm this correlation, using electromotive force (EMF) measurements to determine the thermodynamic activity and electrochemical impedance spectroscopy (EIS) measurements to determine the ionic conductivity of these glasses. Ionic transport was also used to examine the structural relaxation of AgPO3 glass. The glass was heated to another fictive temperature in the glass transition range and its ionic conductivity measured in situ by EIS. Technical Dynamic Composition of Services Driven by User is a solution for developing applications capable of overcoming these challenges. This type of application which will call User-Driven Service Composition Application (UDSCA) allows to compose services during its execution, thus meeting the needs of users. But the lack of guidance on how to develop UDCAs can make it difficult or even impossible to build these applications, because it may aggregate unknown solutions by developers. Looking supply this lack, this work presents an approach to guide developers during the development of this kind of application. To develop the approach, it has been defined which activities should be undertaken during the development as well as the concepts, techniques, artifacts, technologies and tools needed to perform these activities. To evaluate the approach one conducted a case study in which a UDSCA was developed in the field of building maintenance services. The resulting application of the approach was shown to be able to adapt to the heterogeneous needs of the user, also the approach provided artifacts that promoted reuse. Recently, farmers from both settlements were awarded projects of implementation of Agroforestry Systems (AFS) in their plots and the monitoring of these areas, through participatory research, is an important strategy in the regional context. This study aimed to follow the agroecological transition of both settlements through participatory monitoring of forest restoration actions with AFS, seeking to evaluate and understand, through the use of sustainability indicators, changes in socio-cultural, economic and ecological aspects arising from these actions. Therefore, workshops were held with farmers and Participatory Rural Appraisal tools were used for raising awareness and building these indicators. Through participatory methodologies, a set of forty indicators were created with attributes related to soil recovery, socioeconomic aspects, socio-cultural aspects, environment and satisfaction. In a second stage, two monitoring were done; one at the end of the dry season and another at the end of the rainy season, between 2015 and 2016. The monitoring was carried out with the use of various techniques and tools that made possible the participation and understanding of farmers throughout the process. The participatory research included the objectives and the reality of the group, contributing to the perception of farmers for the need of constant monitoring to achieve the sustainability of their Agroforestry Systems. The research brought reflection on autonomy of those involved as to the various needs that the AFS have, revealing the main existing obstacles and difficulties among farmers and helped in the process of appropriation of new management techniques in production systems. The bidding is an administrative procedure in which aims to select, according to predetermined objective criteria, the most advantageous hiring proposal to the government. In this process should be ensured broad participation of stakeholders, as well as equal treatment, always permeated with the legal requirements. Law n. 8.666 / 93, the General Law of Tenders and Administrative Contracts, establishes all the legal system for public procurement. The Differentiated Regime Hiring - DRC - is permeated by several goals, innovative in relation to the rules established by Law 8.666 / 93. UFSCar has adopted this regiment in their hiring process works and engineering services since 2013. The main objective of this work is to promote a comparative study between the Differentiated Regime Signings, DRC and Competition mode, governed by Law n. 8666/93 regarding the efficient procurement of works and engineering services within the UFSCar. For that was used in the methodological procedure descriptive and exploratory research through two approaches: quantitative and qualitative. The first approach corresponded to the comparative analysis, obtained by hiring historical data from the modality competition, the years 2011, 2012 and 2013, and DRC mode the years 2013, 2014 and 2015, coming from the reports of the Purchasing Department - Decom / UFSCar and Comprasgovernamental. They were established some indicators whose purpose was to measure the efficiency of the new procurement regime in relation to the mode set by competition. The qualitative approach was used to analyze the perceptions of representatives of the units that are closely linked to the bidding process under the UFSCar. The main results show that there are many perspectives introduced by the DRC, of which some have already been implemented in the bidding process for procurement of works and engineering services in this federal public education institution. Among the challenges are the various requirements, legal, technical, economic, financial and regularity with which the government by constitutional and legal provisions, is obliged to demand.. It was found that there are many drivers listed in DRC of which is an instrument capable of providing more agile, economical and efficient procurement ways, especially compared to the competition mode. On the other hand, being a relatively new law, the DRC still lacks depth studies that aim to remedy many of the doubts in its various devices and mechanisms. Skeletal muscle ammonia metabolism occurs via synthesis of glutamate and glutamine via critical TCA intermediate α-KG, that regulates increase expression of hypoxia inducible factor 1α (HIF-1α). Furthermore, there is a interaction between HIF-1α and NF-kB. Objective: This study evaluated the effects of hyperammonemia in NF-kB and HIF-1α cross-talking. Methods: To examine the effects of ammonium acetate intervention under HIF-1α signaling through NF-kB pathway it has generated a stable knockdown cell line for NFkB and the subunits α and β of the I kappa B kinase (IKK) complex (IKKα and IKKβ) evaluating the HIF-1α and myostatin activities. Results: The protein expression of HIF- 1α was significantly higher in C2C12 murine myotubes under ammonium acetate intervention compared to control. Once the deletion of NF-kB, IKKα and IKKβ occurs, the HIF-1α is not expressed, suggesting a cross-talking between them. The protein expression of myostatin was significantly higher in C2C12 IKKα deletion under ammonium acetate intervention compared to C2C12 random suggesting that myostatin is not IKKα dependent. This study aims to define a set of performance indicators for the management of technology transfer offices in Brazil, to evaluate, monitor the development and evolution of their actions and accomplishments. Therefore, there was a review of literature on the subject and access to and knowledge of international practices of intellectual property management and technology transfer in universities , which allowed consideration of performance indicators found so far, and the identification other new again featured and updated based on the literature. Were subsequently validated by experts (researchers with publications in the field and qualified professionals who work directly in the management of the NIT) and then there was the application of performance indicators in Brazilian universities (case studies), with reference (for obtaining data) 2015 (alone) and accumulated in the last five years (2011-2015). The importance of the issue is justified by the possibility of using them to understand, assess and monitor the evolution of the university technology transfer process for the company. The study may have great value to understand the actions taken by the NIT, contribute to the formulation of public policies (internal and universities), with the creation of a favorable environment for synergy and cooperation with business, and finally as a process accountability to society (demonstrating the achievements of technological innovation nucleus). In Brazil plantations are concentrated in the Midwest and North, and due to the increase in these exotic plantations, monitoring is necessary to learn about their insect diversity. In 2008 a program to monitor the quarantine pest Sinoxylon unidentatum (Coleoptera: Bostrichidae) in Tectona grandis was started in the state of Pará, Brazil, because of the proximity to the state of Mato Grosso, area of occurrence of this pest. The general objective of this research was to monitor S. unidentatum and other beetle borers in Canaã dos Carajás, Cumaru do Norte, Pau D’ Arco, Rio Maria, São Domingos do Araguaia and Ulianópolis. Our specific objectives were to determine the richness and diversity of Curculionidae (Scolytinae, Platypodinae) and Bostrichidae in these plantations, and to know if there was a seasonal variation for their most abundant species. Trappings were done biweekly with ethanol-baited flight intercept traps from June 2011 to April 2013, totaling 115 collections. We captured 10147 specimens of beetle borers, distributed in eight sub-tribes, and totaling 90 species. In Rio Maria we found the greatest diversity of beetles and in São Domingos do Araguaia the greatest richness. There was a predominance of Scolytinae of the sub-tribe Cryphalina, which probably develop in the understory or dry material above the soil. We trapped significantly more Bostrichidae and Hypothenemus during the dry season. Ngô beyêt is as they call the dam, "standing water, water locked" and expand the meaning of the term for "rotten water, dirty water, old water." In order to strengthen this image of Belo Monte, the thesis presents an ethnographic description of the Xikrin criticism regarding of the white policy in three of its elements: meeting, document, project. The negative elements of the white policy are treated analytically as artifacts being opposed to the forms of existence valued by Xikrin kukràdjà, Mẽbengôkre culture. The contrastive analysis, as well as an important methodological procedure was a widely used resource by Xikrin to express their criticism of the policy of the whites. The contrast images became argumentative form of the thesis, justifying its division into two parts. The analogy by contrast also aims to highlight the image that Xikrin wanted to convey to white of Belo Monte, as a collective of people who share correct and beautiful ways of acting and being in the world. Methods: Sprague-Dawley female rats were grouped into six experimental groups (n = 11 per group): shamoperated sedentary (SHAM Sed), ovariectomized sedentary (OVX Sed), sham-operated resistance training (SHAM RT), ovariectomized resistance training (OVX RT),(OVX Sed-HR), and OVX RT-HR. HR groups received implanted silastic capsules with a 5% solution of 17β-estradiol (50 mg 17β-estradiol/ml of sunflower oil). In a 12-week RT period (27 sessions; 4–9 climbs) the animals climbed a 1.1 m vertical ladder with weights attached to their tails. Biomechanical and physical bone analyses were performed using a universal testing machine, and MMP-2 activity analysis was done by zymography. Results: Bone density and bone mineral content was higher in the RT and HR groups. The MMP-2 activity was higher in the RT and HR groups. The biomechanical analysis (stiffness, fracture load and maximum load) demonstrated better bone tissue quality in the RT associated with HR. It also presents the analysis of the activities undertaken by pupils and conclusions about the proposed objectives and goals achieved. The main objective of the developed activities is to provide students a better learning content covered and issues through a playful approach, interactive and motivating. The specific objectives are to develop the ability to translate a mathematical problem in mathematical language, manipulate algebraic expressions, estimates and comparisons, develop mathematical knowledge as knowing how to express and calculate the area and perimeter of plane figures, calculating probabilities of random events, solve quadratic equations, plotting graphs of quadratic functions and manipulate the software or the GeoGebra application. For this we have created a game of darts adapted and activity sheets containing instructions, questions, tables, graphs, calculation exercises, optimization problems and graphic constructions scripts applied to GeoGebra. The methodology used was the Didactic Engineering. The activities were implemented in two classes of 9th grade of elementary school in two different schools, one class at a school in the municipal school of Bauru, São Paulo, and another class of a school in the state school system the city of Agudos, state of São Paulo. During the application were used 12 50-minute lessons in two classes, with six days of double classes, in which students actively participated in all activities. Our work makes reference to the National Curriculum Parameters (PCN) and other documents governing the teaching of mathematics in public schools in Brazil. We believe in the thesis that ICT can contribute as an educational means for the improvement of teaching and learning situations, and consequently in solving problems such as school failure and school indiscipline. Therefore, we designed and applied a didactic sequence using the Geogebra for the development of Isometries topic in a class of 6th grade of elementary school. The didactic sequence was structured within the perspectives of Didactic Engineering and adopted a qualitative methodology. The type of product to be transported is an important factor in the evaluation of environment-induced cracking, especially of welded joints, where fatigue cracks may originate at points of high stress concentration. In Brazil, the construction of a long distance pipeline has been started in 2014, to transport sugarcane ethanol, which is currently widely mixed with motor fuels. In this work a circumferential joint of an API 5L X70 line pipe was evaluated by hardness, tensile and impact tests; the microstructure was observed by optical microscopy in the different regions of the welded joint and the fractured surfaces were analyzed by SEM. The material of the tube was classified according to API 5L specifications and the welded regions showed hardness distribution and microstructures in agreement with the literature. The fracture surface of the welding metal after tensile and impact tests indicated that the presence of welding discontinuities may affect strongly the fracture behavior, leading to embrittlement, especially under impact testing. The susceptibility of welded joints to stress corrosion cracking (SCC) in a simulated fuel-grade ethanol (SFGE) environment was evaluated by slow strain rate (SSR) and fatigue crack growth (FCG) tests. The tests were designed to promote crack propagation through the heat-affected zone (HAZ) or through the weld metal (WM) of a circumferential weld on an API X70 steel pipe. The SSR tests in SFGE showed reduction of elongation to failure and mixed fracture, with clear signs of SCC in both the regions of the weld. The FCG tests demonstrated that the time-dependence of corrosion fatigue interactions may involve alternating periods of increasing and decreasing crack growth rates throughout the test. Acceleration of crack propagation due to SCC predominated in the WM and deceleration predominated in the HAZ.
