The aim of this research was to investigate the influence of different regions of wood extraction from Angelim Pedra on its physical and mechanical properties. In this research, it was also evaluated, with the help of ANOVA of the regression models, the possibility of being able to estimate the strength properties and some physical properties of the wood as a function of the apparent density. It was concluded that lots from all regions had a high equivalence index; the Bonfim region showed full equivalence and Mucajaí and Claudia showed an equivalence index equal to 93% in both analyses. As for the estimation of properties by bulk density, it was observed that it was impossible to estimate properties by regression models. The characteristic value relationships, used in the simplified characterization of the wood found in this research, were superior to the values established in the Brazilian Standard ABNT NBR 7190; demonstrates that the results obtained in this study can be used, in a new revision of the Standard on the relationships used, for the simplified characterization of wood.listelement.badge.dso-typeItem, Efeito citotóxico e pró-apoptótico do composto [6]-Shogaol contra células de câncer de bexiga (MB49), in vitro(Universidade Federal de São Carlos, 2021-05-24) Nina, Diana Gabriela Nina; Anibal, Fernanda de Freitas; https://lattes.cnpq.br/4918261968772806; https://lattes.cnpq.br/4687877693840586Bladder cancer is considered one of the most aggressive malignancies due to its profile of recurrence and progression, and even with the improvement in methods of diagnosis and treatment, the mortality rate has not shown a declining trend in recent decades worldwide. In this perspective, the search and development of more effective and safer therapeutic alternatives is necessary. Phytochemicals are excellent sources of active ingredients with therapeutic potential, which have been used for centuries for therapeutic and culinary purposes. [6]-Shogaol is a phenolic compound extracted from the rhizome of ginger (Zingiber officinale Roscoe) that has shown antiproliferative and anti-tumor effects in a wide variety of cancer models, both in vitro and in vivo. However, there are no records of studies reporting an antitumor effect in cell models of bladder cancer. Thus, this study aimed to investigate the in vitro cytotoxic and pro-apoptotic potential of [6]-Shogaol against murine bladder cancer urothelial cells (MB49), in relation to non-tumor murine fibroblasts (L929). Through the in vitro study, the cytotoxic and pro-apoptotic effect was evaluated, through the analysis of the reduction of the metabolic activity, observation of morphological alterations associated with cellular damage, identification of alterations in the process of colony formation, evaluation of the level of reactive species of intracellular oxygen, evaluation of alterations in the nuclear structure, identification of apoptosis events and assessment of anti-migratory potential. Based on the results, it was shown that [6]-Shogaol was able to induce cytotoxic effects that compromised cell viability, exhibiting an inhibitory concentration of 50% of cells (IC50) of 135,5 µM for tumor cells MB49 and 205,3 µM for L929 non tumor fibroblasts, in addition to inhibiting and altering the proliferative processes of colony formation, showed pro-apoptotic activity identified by the observation of apoptotic phenotypes, mediated by nuclear fragmentation. In summary, our study suggests that [6]-Shogaol causes cytotoxic damage that compromises the viability of MB49 murine bladder tumor cells in a concentration-dependent manner. This is a descriptive-exploratory study with a qualitative approach. Five respondents participated in the study, with the inclusion criterion working or having worked providing services directly or indirectly to the homeless population by the Municipal Department of Citizenship and Social Assistance or Municipal Health Department. Professional exclusion criteria under the age of 4 years at the Municipal Secretariat of Citizenship and Social Assistance or Municipal Health Secretariat. The municipality has a Consultancy team on street of modality 1, implemented in 2017. Semi-structured interviews were carried out by Google Meet with professionals from the Citizenship and Assistance Secretariat Social and Health for data collection. As a result, two categories emerged: Street Clinic: condition of eternal constrution and Challenges of the intrasectoral and intersectoral network. We concluded that the Street Clinic in the municipality of the study represents an advance in the health issues of people living on the streets, the difficulties of access, prejudices and attempts to articulate with the intra and intersectoral network.listelement.badge.dso-typeItem, Papel da proteína Fbxo7 no perfil de citocinas produzidas em linhagem celular monocítica humana (THP-1) estimulada por lipopolissacarídeo(Universidade Federal de São Carlos, 2021-05-24) Martins, Isabela Fernanda Morales; Teixeira, Felipe Roberti; https://lattes.cnpq.br/3568850159381693; https://lattes.cnpq.br/8976622007212196The canonical function of SCF E3 ubiquitin ligase complexes is the ubiquitination of their substrates, directing them to proteasomal degradation or modulation of function. From these complexes, F-box protein class is the most important component due to their substrate recognition, which ensures the specificity of the complex. In 2012, it was described that Fbxo7 acts as a negative regulator in the nuclear transcription factor kappa B (NF-kB) signaling pathway in human osteosarcoma cell line (U2OS). This pathway is regulated by several ubiquitination events, thus being responsible for regulating inflammation via activation of genes involved in the immune response, including cytokines. So far it has not been evaluated whether Fbxo7 can regulate the cytokine profile through NF-kB. Therefore, the aim of this work was to investigate the role of Fbxo7 in cytokine production using THP-1 cells stimulated by lipopolysaccharide (LPS). Since macrophages help to direct the standard immune response through cytokines, we used the differentiated THP-1 lineage for this study. We performed FBXO7 knockdown by specific siRNA in these cells and stimulated the NF-kB pathway with LPS, followed by the evaluation of 36 cytokines through an array containing capture antibodies for each cytokine. We verified a reduction in the amount of CCL1, CCL3, IL-10 and IL-1β in FBXO7 knockdown cells when compared to the control. Considering that IL-1β is a potent pro-inflammatory cytokine and is produced mostly by macrophages, we also evaluated the expression of the IL-1β gene by RT-qPCR. We noticed that, although there was a reduction in the protein level of IL-1β, there was an increase in transcription of this gene, which may suggest that cells are trying to compensate the reduction in protein levels by increasing gene expression. In addition, we did not find any change in the transcription of NF-kB after the knockdown of FBXO7. Considering that IL-1β is post-translationally regulated, we suggest a regulatory mechanism of Fbxo7 for IL-1β secretion by LPS-stimulated, differentiated THP-1 cells. To that end, we built an appropriate interaction between a qubit, that is, a 13C nuclear spin, and a reservoir, in our case, a large number of nuclear hydrogen spins that act as a spin bath. For this study, we used the carbon-hydrogen structure present in the polycrystalline sample of adamantane. The interaction we seek is built by applying a sequence of specific radiofrequency pulses using the technique of nuclear magnetic resonance (NMR). The temperature of the entire system can be controlled by preparing the initial state of the nuclear hydrogen spins and, thus, we show how to build reservoirs with arbitrary temperatures, including negative effective ones. As an application of these artificial reservoirs, we investigated the implementation of quantum thermal machines. First, we performed an experiment in which a quantum thermal machine works under two reservoirs, one at a positive and the other at an effective negative spin temperature. We use the liquid chloroform molecule to simulate the qubit system. We show that the efficiency of this thermal machine can be greater than the case when the two reservoirs are at positive temperatures. We also demonstrate a counterintuitive result, in which the efficiency of the Otto cycle can be surpassed and when the quantum thermal machine operates in a finite time, that is, in a non-adiabatic manner. Finally, we implemented a quantum thermal machine that operates in a single reservoir at an effective negative temperature whose efficiency is always 100%, regardless of the unitary transformation carried out on the qubit, as long as its final state is different from its initial one. In this experiment we again use the carbon-hydrogen structure present in the polycrystalline sample of adamantane. As far as we know, this is the simplest and most efficient quantum thermal machine ever built. For all steps, we show that the theoretically predicted results are in very good agreement with the experimental data.
