Especially on the northeast stretch, interrupted due to environmental infractions. We investigate and discuss the possibilities and limitations of the conclusion. This study is motivated by the relevance of this road connection, which should interconnect important urban sectors of the city that today remain more isolated. To carry out this research, information was collected and compiled in two main aspects, the urban and environmental characteristics of the affected region and the relevant legislation, especially concerning the urban permanent preservation area (under Brazilian law 12.651) and urban planning instruments that could be applied, which involves urban and environmental requalification with provision for re-establishing the road connection. Based on the data analyses, is possible to formulate alternatives path to complete the northeast perimeter, through alternatives path for connection, which allows the identification of their respective possibilities and limitations. To achieve the research objective, five interviews were conducted from the perspective of oral history with mothers of transgender people who live in the cities of Campinas / SP and Sorocaba / SP and who participate in movements and actions for the rights of trans people. The categories family and transsexuality were analyzed, making an excerpt in their memories of the themes of childhood, education and gender transition. The results showed that the acceptance of these mothers to the transgenderity of their daughters and sons, breaking political, social and religious “norms” that challenged them, are the result of affections, the search for knowledge about gender identity issues and the collective involvement with other mothers of transgender people and with institutions fighting for rights and respect for the LGBTQIA + community (Lesbians, Gays, Bisexuals, Transvestites and Transsexuals, Queers, Intersexes, Asexuals and other groups and variations of sexuality and gender. It is concluded that the five mothers who came out of the closet in favor of the struggle for the legitimacy of the existence of their transgender daughters and sons, came to welcome them and them through a process of constant search to find out about gender and sexuality issues, through references sought in collectives , NGOs and even small groups of mothers of LGBTQIA + people. Childhood, education and gender transition are fundamental categories and n the entire process, as they constitute transgender subjects and subjects. The collectivity of mothers makes a difference in the process of welcoming daughters and sons by these mothers. The problem-posing question covers the following inquiry: how do the toys made by miriti artisans in Brazil carry the cultural identity of those who produced them? When it comes to the methodology used, the research has a qualitative approach, as to its general objectives, it is of the exploratory type and, in terms of the nature of the collected sources, it is bibliographic. Unfortunately, with the current scenario impacted by the Covid-19 pandemic, it was not possible to use physical libraries, cultural centers or artisanal toy fairs, so the research for this paper was made mostly via digital means. In order to clarify this question, the cultural history of the toy was initially addressed, as well as its industrialization process - and the resulting impact on toy production and the relationship established with children - and the toy as a cultural support. An overview of the handicraft production in Brazil was also presented, followed by an explanation about the miriti toys regarding the history and production of these items within the abaetetubense community. Finally, the richness and potential of handcrafted toys in comparison to industrialized toys were reiterated, and the importance of valuing this type of manual production was emphasized.listelement.badge.dso-typeItem, Predição de proteínas parceiras de interação da trealase, enzima relacionada com a patogenicidade de Xanthomonas citri subsp. citri(Universidade Federal de São Carlos, 2021-06-17) Brambila, Beatriz; Novo-Mansur, Maria Teresa Marques; https://lattes.cnpq.br/1198926223396748; https://lattes.cnpq.br/2803161066730904Known as one of the main phytopathologies that affect the citrus industry worldwide, citrus canker is caused by the bacterium Xanthomonas citri subsp. citri (XAC) and is responsible for causing premature drop and reduction in fruit quality, directly affecting productivity and causing a relevant economic impact. Through previous proteomic analysis of our research group, it was possible to identify the periplasmic trehalase enzyme (encoded by the XAC 0604 gene) as a protein expressed in XAC when cultivated in pathogenicity-inducing medium (XAM-M). For the purpose of functional studies, a mutant strain deleted in the gene encoding the trehalase enzyme (XACΔ0604) was previously obtained by our group, as well as the construction of the vector to obtain the recombinant trehalase protein by heterologous expression in E. coli. Considering the scarcity of works in the literature about the functionality of this enzyme, especially in XAC, the present work aimed to verify if there is an interaction between trehalase and other cellular proteins and if there is a change in the interaction profile when using the mutant strain lysate (XACΔ0604), compared to the wild strain (XAC). Thus, the recombinant trehalase was immobilized in a column for a pull-down experiment (in vitro), followed by the addition of cell lysate from wild (XAC306) or mutant (XACΔ0604) strains, cultivated in duplicate (A and B) in pathogenicity-inducing medium (XAM-M). After successive washing steps, the complex was eluted from the column with 250 mM and 2 M imidazole. Analysis of the profiles of the constituent proteins of the eluate was performed by SDS-PAGE, the most evident bands were isolated and digested using trypsin for further analysis by mass spectrometry. For the wild strain, bands that co-eluted with the expected size band for trehalase (~61kDa) for replica A were observed, with approximate molecular mass values of 51, 46, 27 and 19 kDa, while for the replica B, the bands that stood out were those with values of 51, 46 and 27 kDa. The eluate obtained with the mutant strain lysate showed bands of 51 and 27 kDa by SDS-PAGE, similar for both cultures A and B (replicas), in addition to the band predicted for trehalase. Thus, the 51 and 27 kDa bands were recurrent between the two strains. The identification step by mass spectrometry of the proteins present in the isolated bands was made impossible due to the interruption of LNBio-Campinas activities during the COVID-19 pandemic, which was an important setback for the completion of the present work. Thus, we performed the protein-protein interaction prediction by the STRING bioinformatics tool, where it could be foreseen that a likely interaction partner of trehase in XAC is the protein trehalose-6-phosphate phosphatase (OtsB), involved in trehalose biosynthesis, which has a molecular mass of 27.0 kDa, which corresponds to the mass of one of the recurrent bands identified by the pull-down both in the wild and in the mutant. To understand the experiences of women in childbirth and postpartum and their associations in maternal role attainment. This is a quantitative-qualitative, exploratory and descriptive study, based on two steps, application of the Mother-Baby Friendly Birthing Facilities (MBFBF) Thermometer and semi-structured interview with women in the first stage. The data collected in the first stage (quantitative) were obtained through a convenience sample and analyzed descriptively; while in the second stage (qualitative), the data were interpreted by content analysis in the thematic modality addressed by Bardin. The results of the first stage reveal that the women assisted in the institution related to the study are not free of harm. The statements of the women obtained in the second stage of the study resulted in the construction of three thematic categories: 1. "Previous knowledge: power to give birth and care", 2. "Professional posture and becoming a mother" and 3. "Importance of the support network and the pandemic in maternal role attainment ". The experience of women is surrounded by intense feelings, from tension and fear to complete satisfaction and the joy experienced in a unique and transformative moment, which they report with great richness of detail through a look at the posture of each professional who assists them, as a way of facilitating this transition to motherhood. For instance, virusestransmitted by arthropod vectors can widely spread in wild landscapes and in urban areas. This is the case of Oropouche virus. This emerging arbovirus can be transmitted in urban areas through Culicoides paraensis’ prick. The Culicoides paraensis is a hematophagous midge, which circulates in Brazilian north and northeast. The Oropouche virus cause an exanthematic febrile illness in humans, which in some cases, can evolve to neurological complications. Although, the pathogenetics details and the role of key transcription factors of the innate immune response on control of this viral infection are not completely understood. Furthermore, IRF-5, a transcription factor activated after the recognition of pathogen-associate molecular patterns, is essential to control neuroinvasion by Oropouche virus. Thus, the aim of this study was to characterize how the absence of this gene in essential cells of the immunological response, such as macrophages and B lymphocytes, could affect the disease progression caused by Oropouche in a murine model. To achieve this goal, a recombination system known as Cre Lox has been taken. As a result, the virus has achieved the central nervous system of both lineages. Oropouche has caused meningitis, encephalitis and conditional mutants’ death. Highlighting the IRF5’s importance in these leucocytes in front of neuropathogenesis induced by this virus. In conclusion, there is a requirement to investigate how Oropouche virus gain access to Central Nervous System. From this context, this research aimed to analyze the Reading and Writing Program and its implementation in the state school system of São Carlos/SP, verifying its implications in the teaching work, based on the evaluation of teachers about this policy. As specific objectives, this study aimed to: understand the historical and political context of the Reading and Writing Program; analyze the structure and functioning of the Program, verifying, mainly in its materials, the relationship it maintains with teachers; identify whether teachers use reading and writing program materials in their daily activities, why and how; to investigate the contributions and limitations of the Reading and Writing Program in the development of teaching work, according to the teachers' analysis. To achieve the proposed objectives, bibliographic, documentary, and field research were used. Initially, a bibliographic and documentary analysis of the theme was made, with the intention of mapping the object of this research, in addition to studying its structure and functioning, as well as its history - whose theoretical field led to the study of the New Public Management, a new form of management of educational policies that have been incorporated in the State of São Paulo since the mid 1995s. Furthermore, this research also went through an empirical stage, using the on-line questionnaire as an instrument of data collection - sent to the teachers of the São Carlos state network, using the “Microsoft Office Forms” program. This stage obtained 24 collaborators, who agreed to respond to the survey. For data analysis and collection, this work used content categories, according to the perspective proposed by Kuenzer (1998). Categories were defined, in this sense: teachers' understanding of the material; daily teaching work and frequency/use of the Program materials; difficulties/limitations regarding the material and impact on teaching work; contributions for the use of materials to the teacher; teacher training. With this research, some results were reached, among them: teachers understand the materials of Reading and Write positively, but do not consider them sufficient for the reality lived in state schools, since they are out of the singularities existing in each context; they mention that there are difficulties in working with materials and that the limitations are to treat schools, teachers, and students in a standardized way; understand that the Program acts for the construction of a dependent teacher, who does not perceive himself capable and sufficient to create solutions to the reality itself; nevertheless, they also show that they often use the materials proposed by the Program, by making sense and meaning in each classroom, adapting them to the realities existing in schools. Thus, what is observed is that the Read and Write Program has contributed little to the quality of São Paulo's education, even less so, to the strengthening, care, and autonomy of teaching work. In Civil Engineering, great changes have been taking place and it is important to raise a panorama of these new technologies in order to stimulate the interest, creation, adoption and development of these technological innovations. Building maintenance is a branch of civil construction that has considerable space for evolution in its studies and innovations. The phase of use and operation is the longest and bigger cost in the life cycle of a building, so the maintenance management requires methods and tools to assist this process, such as the use of BIM, drones, sensors, among others. The present work seeks to gather tools, technologies and methods characteristic and coming from industry 4.0 used in the management of building maintenance. For such used a methodology of research, extraction, processing and analysis of bibliographical data of articles, theories, among others, known as RBS (Systematic Bibliographical Revision), which aims to compile and to create analytical basement on the subject. The intention of the research is to favor each time more the use and applications of these techniques, since the results of their uses involve increase of productivity, quality, precision and reduction of deadlines. Nowadays, it’s known that some damaged seeds can still germinate, and grow as seedlings. In this study we used seeds from Eugenia pyriformis, E. involucrata and E. uniflora to: (i) to test what is the threshold of damage the seeds can tolerate and if tolerance is linked with seed size; (ii) to evaluate the viability of these seedlings, considering the gap of information about the late effects of partial predation; (iii) to investigate if seed damage affect resource allocation to the roots or stem. Seeds were damaged to simulate the partial consumption by a rodent. The seedlings were planted in tubes and kept in a greenhouse for three months. Seedlings emerging from large seeds, represented by E. pyriformis, were not affected by light damages, and light damages stimulated seedling growth. Seedlings from small seeded plants were more affected by damages, resulting in smaller seedlings. Seedlings from E. uniflora invested more in root growth, while E.involucrata invested more in stem growth. Over the years, with the modernization and professionalization of campaigns, political parties have adapted to the new reality, hired marketing professionals, and adjusted their strategies and thematic agendas to the communicational logic. In Brazil, this process is observed mainly in the campaigns broadcasted in the Horário Gratuito de Propaganda Eleitoral (Free Time for Electoral Propaganda). Television was for many years the main source of political information, and thus HGPE is a privileged space for the presentation and observation of themes. For 20 years the Brazilian campaigns were polarized between PT and PSDB, together these parties obtained the largest part of the valid votes and the greatest access to block advertising time. Based on these affirmations, this paper seeks to answer the following questions: Which themes have predominated in the TV presidential campaigns of PT and PSDB over time? Being the hypotheses: 1) There is a decline in the thematic segments presented in HGPE of the presidential disputes between PT and PSDB between 1994 and 2014. 2) Due to the professionalization of the campaigns, the parties present a thematic convergence over the years 3) Even though the parties present theme convergence over the years, the content of the themes presented is divergent. The results show that the parties do present a convergence in the thematic agendas, but that this convergence is not expressed in the strategies used to present them. In addition to the edge effects caused by habitat fragmentation, its plant and animal species also face the effects of climate change. Such changes can affect climate variations and cause more frequent extreme events, such as droughts. Plants and animals respond to abiotic variables and such responses are important to understand the effects of climate change on organisms and their interactions. Palm trees are an important element within the Atlantic Forest, mainly because they produce fruit during the dry season of the year when there is less supply of resources for fruit-eating animals. Plant-animal interactions can be beneficial to plants, such as seed dispersal, or negative, such as seed predation. Thus, understanding the factors that somehow impact the reproduction of plants that provide resources for other organisms is essential, since changes in reproductive characteristics of an organism can be reflected for other species, such as its predators. In this work we evaluated whether droughts alter the reproductive phenology of Syagrus romanzoffiana, a key species in the Atlantic Forest, and whether the possible alterations have cascading consequences for animals that consume its seeds. The main results indicate that drought increases the number of bunches of immature fruits, but reduces the number of bunches of flowers and ripe fruits. Droughts can also delay or advance the time of occurrence of these structures. However, such changes do not reflect differences in predation rates of Syagrus romanzoffiana, whose main predator (beetle) is not satiated by increased seed production, unlike the vertebrate predator, squirrels. In the optimization process, restrictions on decision variables are determined, according to which continuous values can be accepted, which constitute a linear programming problem. In a general problem of integer linear programming, we seek to minimize a linear cost function over all n-dimensional vectors x subject to a set of linear restrictions of equality and inequality, as well as integrality restrictions in some or all variables in x. If only some of the variables are restricted to take integer values, and others can take real values, then the problem is called a mixed integer linear programming problem (MILP). If the objective function and/or the constraints are nonlinear functions, the problem is called a mixed integer nonlinear programming problem (MINLP). If all variables are restricted to assume integer values, then the problem is called a pure integer linear programming problem. If all variables are restricted to assuming binary values (0 or 1), then the problem is called a binary optimization problem, which is a special case of a problem of pure integer linear programming. The branch and bound method is not a solution technique specifically limited to integer programming problems. It is a solution approach that can be applied to many different types of problems. The branch and boundary approach is based on the principle that the total set of viable solutions can be divided into smaller subsets of solutions. These smaller subsets can then be evaluated systematically until the best solution is found.
