Item type:Item, Desenvolvimento de procedimentos automáticos para a determinação de 3-hidroxibutirato, glicose e colesterol em soro de sangue animal empregando multicomutação em fluxo(Universidade Federal de São Carlos, 2003-09-23) ;Pires, Cherrine Kelce ;Reis, Boaventura Freire doshttps://lattes.cnpq.br/7619359855870896In the present work, in flow injection systems were developed for the determination of important metabolic parameters, such as 3-hydroxybutyrate, glucose and cholesterol in animal blood serum. The analysis modules were developed based on the multicommutation concept, whose main characteristics favor the processing of many samples. For this, three-way solenoid valves were used, as discreet commutation devices, for the intermittent addition of samples and reagents, controlled by a microcomputer equipped with a commercial electronic interface (PCL-711S). The control and data acquisition programs were written in Quick BASIC 4.5. The proposed methods are based on enzymatic reactions. For these, the enzymes were immobilized in glass beads and packed in mini-columns of acrylic (15 x 5 mm i.d.). The determination of 3-hydroxybutyrate was based on the reduction of nicotinamide adenine dinucleotide (NAD+), forming NADH, which was monitored at 340 nm. The analytical curve of the proposed system presented a linear dependence on concentration for concentrations between 25 and 150 mg L-1, an analytical frequency of 60 determinations per hour, relative standard deviation of 1.4% (n=17), estimated for a sample containing 75 mg L-1 of 3-hydroxybutyrate, and a detection limit of 2 mg L-1. Other observed favorable aspects were low reagent consumption of NAD+ (0.9 mg) and 3-hydroxybutyrate dehydrogenase (8 µg), and 200 µL of sample per determination. Another analysis module was developed for glucose determination using chemiluminescence detection. The glucose was oxidized by glucose oxidase to gluconic acid and hydrogen peroxide. The hydrogen peroxide formed reacted with the luminol, in the presence of the catalyst hexacyanoferrate (III), producing a blue luminescence at a wavelength around 420 nm. The analytical curve of the proposed system was linear for concentrations between 50 and 600 mg L-1, showed a relative standard deviation <3.0% (n=20) for a sample containing 300 mg L-1 of glucose, a detection limit of 14.0 mg L-1, and an analytical frequency of 60 determinations per hour. The sample, hexacyanoferrate (III) and luminol consumption was 46 µL, 10.0 mg and 0.2 mg per determination, respectively. To conclude this work, the determination of cholesterol was proposed. The esters of cholesterol were hydrolyzed by cholesterol esterase to free cholesterol and fatty acid. The free cholesterol was oxidized by cholesterol oxidase to cholestenone and hydrogen peroxide. The hydrogen peroxide reacted with luminol and hexacyanoferrate (III), being detected by chemiluminescence. The proposed system presented an analytic frequency of 40 determinations per hour, relative standard deviation of 2.3% (n=20) for a sample containing 75 mg L-1 of cholesterol, a linear dependence in the concentration range of 25 and 125 mg L-1, and an estimated detection limit of 3.7 mg L-1. The reagent consumption was 0.22 mg of luminol and 2.75 mg of hexacyanoferrate (III) per determination. Once the best analysis conditions were found, a set of samples was analyzed using the three proposed systems. In view of recent contributions of scientific literature in the educational domain regarding the centrality of the educator in the formative process, considering also its subjectivity in association with the available objective conditions (such as didactic resources, institutional culture etc.), this work aims at the perspectives of projects such the above mentioned one and is focused on answering to the question: "what conceptions, yearnings and expectations are present, under the optics of its individuals, in schools that are in the imminentcy of the implantation of technological innovation projects ?" Having as its main data source the reports "collected by means of half-structuralized interviews" directed to the individuals more directly involved in the SFP, the results show differences on the individual s degrees of involvement, which are in proportional relation to the institutional hierarchic thereby highlighting the importance of "(re)thinking" about the school structure while a community that teaches and that learns. Also it as verified the concurrency between the new challenges considered for the innovative project and the old difficulties that characterize the school (fragmentation and overload of both the functional and the organizational structure of the school) what it seems to be aggravated by the vision of the different individuals regarding the historically instituted relations between the school's internal and external instances (understanding these last ones as the educational agencies and politics). Moreover, some "veteran questions" of the educational universe seem to re-emerge driven by the feeling of instability generated by a technological innovation project within the school, namely: the relation school/family/community, curricular election and contents teaching, skills and professional development of the educator, discourses and reductionist practices of the pedagogical act. Concluding, we consider the importance of the school in the transformation vertex intended for a faired and more humanized society, and also propose some perspectives so that initiatives such as the School of the Future Project can be effective by overcoming ruptures among specificities, attributions and instances involved in projects carried on within schools, as well as overcoming of the trends toward the emphasis on the pedagogical seduction, on the undefinitions, instabilities and resistances against the necessary rearrangements in the organizational and functional structure of the school (culture, subjectivities, spaces, times, demands, "customers") and, finally, on a existential crisis of the school about "why and how" to educate and to educate themselves.Item type:Item, Efeitos da prolactina sobre a massa óssea de ratas ovariectomizadas(Universidade Federal de São Carlos, 2003-09-26) ;Driusso, Patricia ;Nonaka, Keico Okinohttps://lattes.cnpq.br/4551036775738296Prolactin (PRL) has a great number of biologic functions in many species. Beyond the well known effects on reproduction, lactation and maternal behavior, many studies have showed the effects of PRL on the mineral transportation, calcium metabolism and bone. PRL seems to have an effect on the bone, acting direct or indirectly. The aim of this study was to investigate the effects of the PRL on bone mass of female ovariectomized (ovx) Wistar rats, through physical, biomechanical and biochemical analysis of the femur, biochemical of serum and densitometry of the tibia. The rats had 4 months of age in the beginning of the study and were divided in 3 groups: basal group, hiperPRL group (intact, ovx+sham e ovx + 2 grafted anterior pituitary under the kidney capsule) and hipoPRL group (intact+saline, ovx+saline and ovx rats treated with bromocriptina-3 mg/Kg). Statistical analysis of results was performed by a two way analysis of variance (ANOVA) followed by a Student Duncan test. The ovx promoted the figher weight gain and bone growth. It was observed that the ovx+hipófise group showed a significant decreased in maximum force and stiffness when compared with the intact group, and a significant decreased in the densitometry, mineral bone density and percentage of mineral material of the femur when compared with the ovx+sham group. The ovx+ bromocriptine group showed a significant increase in maximum force, stiffness and bone volume of the right femur and the bone mineral content of the tibia when compared with the ovx+saline and basal groups. The work was conducted in commercial grow-out ponds located adjacent to two estuaries in the Eastern coast of the State of Ceará, Brazil. One semi-intensive production cycle was monitored throughout the dry season. In the wet season of 2002, three production cycles (two semi-intensive and one super-intensive) were studied. Environmental conditions and management techniques adopted were characterized through tabular and descriptive analyses. Microbiological conditions in the sediment and water of the farmed and natural ecosystems, as well as in cultured shrimp were studied through standard plate counting using PCA and TCBS media. Presumptive bacterial colonies of the genus Vibriowere isolated and identified. A general evaluation of shrimp health status was conducted through wet mounting and histopathology of shrimp organs. Final shrimp grow-out data indicated management practices and environmental conditions positively influenced the balance between the two ecosystems. The type of rearing system used did not influence the number of bacterial present in each ecosystem and in farmed shrimp. Also, the incidence of bacterial and (or)virus lesions in shrimp was not related to the level of intensification. Changes in shrimp health conditions as observed by wet mount analysis were in most cases related to farming conditions. This confirmed management techniques are also appropriate to the preventive sanitary management in shrimp farming. According to the results obtained in commercial ponds, it was concluded the occurrence of diseases is not related to the type of rearing system adopted by the shrimp aquaculture operation. However management practices and environmental conditions, reflected in the adoption of best management practices in each grow-out cycle, are vital to the health status of each pond's ecosystem.Item type:Item, Estudo da purificação do àcido clavulânico utilizando processo contínuo de adsorsão(Universidade Federal de São Carlos, 2003-10-02) ;Almeida, Renata Maria Rosas GarciaHokka, Carlos OsamuClavulanic acid (CA), a beta-lactam antibiotic, has been known for its commercial and therapeutic importance. This antibiotic is produced by an actinomicete, Streptomyces clavuligerus, and has a combined use with penicillins, acting as inhibitor of beta-lactamase enzymes. Due this importance, some studies on clavulanic acid production has been developed at DEQ-UFSCar. As a natural consequence of these studies and the global knowledge of this process it was necessary to develop a work studying clavulanic acid purification. The adsorption process was chosen to be studied as the process of clavulanic acid recovery and purification. In the present work a continuous adsorption process for clavulanic acid purification was proposed. This process utilized two stirred tanks (CSTR) with recycle of the adsorbent material. The adsorbent chosen to this process was the ion exchange resin Amberlite IRA 400 in the chloride cycle. First, it was necessary to study the adsorption and desorption kinetics and the clavulanic acid equilibrium isotherms in that resin to investigate the influence of temperature, pH and clavulanic acid initial concentration. The degradation rate of clavulanic acid in basic and acidic solutions is very high; high temperatures also increase the degradation rate. Consequently, studies on clavulanic acid degradation were carried out in the same temperature and pH conditions of the kinetics studies. A mathematical model was proposed to describe the adsorption and desorption processes and the kinetics and transport parameters were determined. The continuous process was also mathematically modeled to obtain preliminary data to carry out the clavulanic acid continuous experiments. The parameters utilized in the continuous adsorption process were optimized by response surface analysis to maximize yield and concentration and purification factors. Experiments of the continuous adsorption process were carried out in some different conditions. The continuous process like proposed was operationally viable, but the concentration and purification factors obtained were lesser than those obtained in fixedbed experiments.Item type:Item, Magnetismo a temperatura ambiente em polímeros derivados de politiofeno sintetizados eletroquimicamente(Universidade Federal de São Carlos, 2003-10-08) ;Forner, Alessandra Alves Correa ;Souza, Ernesto Chaves Pereira dehttps://lattes.cnpq.br/1505400360366643In this thesis two polymers obtained from alkyl derivatives of thiophene, poly(3-methylthiophene) and poly(3-hexylthiophene) were studied. The effects of preparation variables on their electrochemical properties using the factorial design were analyzed. The preparation variables investigated were the supporting electrolyte and the synthesis applied potential. Then, a factorial design 22 was performed for each polymer and was made too a third factorial design 22, comparing the monomer and the potential. All the electrochemical analyses were made to understand the polymer behavior as a first step for the magnetic study. The magnetic properties of poly(3-methylthiophene) were analyzed and the weak ferromagnetic phase was observed in their pressed pellets as the polymer was partially doped with ClO4 - anions. This magnetic behavior was observed in full range from 1.8 K to 300 K with measurements performed using a SQUID magnetometer. The magnetization depends on the pressure used to prepare the pellets and the water content of the solvent used for the synthesis. The intrinsic character of this behavior was shown from atomic absorption analysis that discarded contamination. It was observed that the polymer shows different magnetic phases: diamagnetic, paramagnetic, ferromagnetic, antiferromagnetic and metamagnetic.Item type:Item, Instituição militar e política, uma abordagem antropológica (1964-1974)(Universidade Federal de São Carlos, 2003-10-09) ;Frederici, Sandra MariaLeirner, Piero de CamargoThe purpose of this dissertation is thinking the relationship between the military institution and politics in the period between 1964-1974, by an anthropological reading of the processes occurred in the core of the military hierarchy. Therefore, its analysis deals with the military careers constructed in this period, as well as the intramilitary political arrangements, as fields able to better evidence the movements in the command sequence under the new position assumed in the political context. Small-scale commercial fishermen from Upper-Medium São Francisco River, object of this study, are influenced by the dynamics of the environment cycle and by the ecology and biology of fishery resources, maintaining a very close relationship with them, for developing new knowledge and understandings which can provide better conditions of survive. More than that, the small-scale fishermen have self-organized for planning and practicing local management of fishery resources, which include local rules or institutions which define the rights and duties in the access and use of resources. The study of local ecological knowledge and common property systems developed by these communities are the mainly goal of ethnoecology science and of researches in common property systems, which support them as alternatives to the traditional scientific ecology and management of natural resources. This research was carried out between 1999 and 2001 with 7 field trips of 15 days each. Open and structured interviews were carried out, complemented by direct observation of the fishing activity, and the collect of fishery yield. The small-scale fishermen of Upper-Medium São Francisco River have demonstrated to pursue a detailed comprehension about the ecology and biology of the aquatic system and its fishery resources, on which they have based their fishery practices and common property systems, expecting to guarantee efficiency with sustainability. The experiments described the decomposition in function of temperature, availability of oxygen and carbon source. Samples of this macrophyte and water were collected in the oleo lagoon; incubations were prepared and maintained in controlled conditions. The results allow us to conclude that: (i) the U. breviscapa detritus are a heterogeneous substrata and presented two fractions: a labile/soluble (COPLS » 23.5%) and a refractory (COPR » 76.5%). The mass losses coefficients of the labile/soluble fractions (kT = 1.5 day-1) were higher than the refractory fractions (309 times in the aerobic condition and 469 times in the anaerobic condition). The increasing temperature and the dissolved oxygen concentrations favored the mineralization of the refractory fractions. The COPR mineralization in the aerobic conditions was faster than in the anaerobic conditions (1.5 times); (ii) the enzymatic activities of the cellulase presented a temporal pattern that was affected by the temperature, availability of oxygen and carbon source; (iii) stoichiometric ratio (O/C) varied in function of temperature and time; (iv) the oxygen uptake coefficients (kd) presented low variation in function of increasing temperature; (v) the increment on temperature favored the CH4 formation in the anaerobic decomposition of U. breviscapa and its emissions tended to occur earlier; (vi) probably, the enzymatic degradation of the cellulose in the anaerobic strata of sediments contributed to methanogenesis in the oleo lagoon; (vii) the simulations indicated that heterotrophic potentials of the oleo lagoon presents low variations during the year; the favorable period for detritus cycling is in the spring (October - November), in this period increments in mineralization occurs as function of increasing temperature and high availability of oxygen.Item type:Item, Efeitos de interações elétron-elétron e spin-órbita nas propriedades magneto-eletrônicas de magneto-transporte de sistemas confinados(Universidade Federal de São Carlos, 2003-10-10) ;Destefani, Carlos FernandoMarques, Gilmar EugenioEffects of the direct and exchange electron-electron interaction, external magnetic field, symmetry of the charge carriers confining potential, radius, material g-factor, and also of the spin-orbit interaction in zincblende structure materials, are treated on the electronic and transport properties of semiconductor quantum dots (islands) charged by many particles. Three distinct kinds of confining potentials are considered: spherical, parabolic, and quasi-one-dimensional which, respectively, define a three-dimensional, two-dimensional, and one-dimensional island; the first one is more appropriated for the description of quantum dots formed in glassy matrices, while the last two better describe quantum dots litographically defined in a two-dimensional electron gas. Transport properties are considered in the spherical and quasi-one-dimensional islands, where we assume that the electronic current is in the resonant tunneling ballistic and coherent regimes, with essential role played by the excited states of the specific symmetry. We show that different geometries induce distinct level ordering in the island and that there is, in addition to the usual spin blockade, another kind of blockade mechanism which influences the system current; we label it by orbital blockade, because it is essentially due to the structure geometric confinement. We calculate the electronic spectrum of the many-particle system according to its symmetry. In the spherical case, we firstly use the LS-coupling scheme in order to obtain the eigenstates of an island charged by 3 electrons, following the orbital L and spin S total angular momentum addiction rules; we consider intensities of the magnetic field that allow us to neglect its diamagnetic contribution; the electron-electron interaction is treated as a perturbation in a Hartree-Fock way. In the following we use, in this same symmetry, the Roothaan and Pople-Nesbet matrix methods in order to deal with islands charged by 40 electrons, where the addition spectrum is calculated and Hund s rule is verified; we show how a magnetic field is able to violate such rule. The advantage of this numerical approach is the possibility to deal with a very high occupation in the island; the disadvantage is that their eigenstates do not have defined L and S values, as it is the case in the LS-coupling scheme. In the parabolic case, we employ a numerical diagonalization in order to obtain the island eigenstates charged by 2 electrons, without any restrictions regarding the magnetic field intensity or the system radius; we take into account both possible spinorbit couplings, one related to the implicit absence of zincblende crystalline structure inversion symmetry (Dresselhaus effect), and the other one related to the absence of structure inversion symmetry as caused by the confinement defining the two-dimensional electron gas (Rashba effect); we analyze the critical magnetic fields where both effects give origin to a intrinsic spin mixture in the island, inducing level anticrossings in the Fock-Darwin spectrum where intense spin-flips processes occur. In the quasi-onedimensional case, we just reproduce a known spectrum for an island charged by 4 electrons.Item type:Item, Abordagem estatística e computacional na otimização de meios de cultura para produção de antibióticos: o caso da cefalosporina C(Universidade Federal de São Carlos, 2003-10-10) ;Ferreira, Amauri AlvesSuazo, Cláudio Alberto TorresThe scope of the current work was to investigate the effect of glucose, amonium acetate, DL-methionine, phosphorus, oleic acid and salt solution on the maximum specific growth rate (µmax), as well as the effect of sucrose and DL-methionine on the specific cefalosporin C production rate (µp), by Cephalosporium acremonium ATCC 48272, using factorial design and response surface techniques. The initial medium of fermentation was used according to factorial design and the experiments were carried out in shaker flasks at controlled temperature at 28 °C, initial pH of 7,0+0,1 and agitation speed 250 rpm. For both specific rate the variables involved were correlated by a fitted second order model. The canonic and surface response analysis showed a maximum specific growth rate of 0.22 h-1 obtained when glucose and amonium acetate concentration, were 4.2 , 2.5 g/L, respectively, and specific cefalosporin C production rate of 1.12 mgCPC/gcel.h when sucrose and DL-methionine concentration, were 10.5, 2.5 g/L, respectively, for a celular concentration of 14.0 g/L. The experimental design showed to be very efficcient in evaluating the bioprocess, as long as high values of µmax e µp were obtained in comparison with literature data. The feedforward neural network (FNN) was employed for modelling and simulation the effect of glucose, amonium acetate, DL-methionine, phosphorus, oleic acid and salt solution on the maximum specific growth rate. The network topologies were used with 6 neurons in the input layer, 22 hidden layer and 1 output layer. The data base for training the FNN was obtained from results of the factorial designs processed with a backpropagation learning algorithm. The surfaces response obtained by simulations for all range of glucose and amonium acetate concentrations showed inhibitory effect for the two substrates on the maximum specific growth rate. The results of the simulation are in good agreement with the experimental and statistical model results.Item type:Item, Ictoplâncton do complexo estuarino de Itapessoca norte de Pernambuco - PE(Universidade Federal de São Carlos, 2003-10-13) ;Souza, Rosana Coimbra ;Verani, Nelsy Fenerichhttps://lattes.cnpq.br/1862746100743391The present study was carried out in the estuarine complex of Itapessoca, located on the north coast of Pernambuco (PE) Brazil, at latitudes 07º 37 - 07º 41 S and the longitudes 034º 50 - 34º 55 W, from April 2001 to April 2002. The purpose was to characterize the environment and the ictioplanktonic community, attempting to recognize the distributional trends of these organisms, in order to evaluate the estuary s role as a site for the fish initial development phases. The water temperature, salinity and transparency were measured and the collected samples of the water where analyzed in order to estimate the levels of nitrite, nitrate and dissolved phosphate, ammonia, alkalinity and chlorophyll-a. The ictioplankton was collected through horizontal hauls with plankton net with 500 µm mesh. The hauls had a standard duration of 10 minutes, with the approximate speed of two knots. It was calculated the specific diversity of the fish larvae and the biotic and abiotic data were statistically related. A total of 87,758 eggs and 6,677 larvae were sampled corresponding to a total density of 39,445.34 eggs.100m-3 and 3,001.17 larvae.100m-3. The eggs and the larvae were present in all months and sampling stations. In relation to the fish larvae a total of 8 orders and 19 families were identified. The Engraulidae family was the most abundant (69,53%), followed by Eleotridae (7,73%) and Gobiidae (6,53%). Despite of the considerable integrity state of the Itapessoca estuary, the values of the diversity family indexes were low, in comparing to other estuarine areas. These low values can be explained by the dominance of the Engraulidae family, in all months and sampling stations. It was verified that the environment is rich in phosphate, with high levels of alkalinity; the levels of chlorophyll-a were in accordance with productive environments, but the low values of nitrite, nitrate and ammonia can indicate that nitrogen is a limiting factor of the local productivity, reflecting in the low occurrence levels of ictioplanktonic organisms at the estuarine complex of Itapessoca.Item type:Item, Proposta de uma metodologia baseada em indicadores de desempenho para avaliação de principios ralativos à produção enxuta: estudo de caso em uma empresa fabricante de produtos para o setor médico-hospitalar.(Universidade Federal de São Carlos, 2003-10-15) ;Dias, Flávio TeodoroFernandes, Flavio Cesar FariaThe objective of this project is to develop a methodology to evaluate the performance of a production system through the use of performance indicators regarding lean production in relation to the objectives of quality, manufacturing and productivity and its variables, quality, time, and cost. A presentation of the evolution of the lean concept will be performed, its principles and techniques, and the presentation of the three views of the lean thought. Within this context, a new view will be presented which seeks a formal integration between the production control and quality control at the shop floor level. The most used performance indicators will be described. They are used to measure the performance of a lean system and levels of employment for each indicator will be defined at the levels of the shop floor, company and supply chain. This project proposes a methodology that allows measurement of a company s performance that quantifies benefits that can be expected in the implementation of lean techniques. Finally, it will be shown through the application of the proposed methodology in the case study that the introduction of the concepts discussed in the fourth approach of lean production, which deals with the change of material and information flow from the production systems, helps the company to be more competitive. The target public for this study were fifth grades students attending the state public school Antônio Militão de Lima and their respective teachers. In order to achieve the proposed objectives, a pedagogical intervention which privileged the Theory of Social Representation was established, based on the methodology developed by the educator Marcos Reigota and an interdisciplinary program, whose aim was to arouse the elements involved towards the forming of citizenship and ecological awareness. The teachers were oriented to the importance of collective and interdisciplinary involvement through offering capacitating courses and interaction with local reality. The results obtained demonstrated the true difficulties faced today by the Public Scholl, and their development of program allowed us to conclude that: 1) capacitating courses for teachers could contribute to arousing a mere critical vision of reality in the educator; 2) the results brought a series of information (informal) through interviews as well as through classroom discussions regarding the children s learning. Main features in DS include facial and dermatological features, congenital heart defects as well as immunological, gastrointestinal and endocrine abnormalities. Among a variety of cancer, a 20 fold increased risk of developing leukemia in younger people and an increased risk of testicular cancer is noticeable in DS patients. Most DS patients carry a complete trisomy of chromosome 21, but patients carrying a partial trisomy have also been observed. Analyses of partial trisomy of chromosome 21 have helped determining the minimal regions potentially associated with DS features. The DSCR2 gene located in the so-called Down Syndrome Critical Region 2 (DCR-2) codes a 32.8-kDa leucine-rich protein comprised of 34% hydrophobic amino acid residues. Little is known about the DSCR2 protein characteristics but many assumptions have been made according to in silico predictions. In this work, we used immunocytochemical and fluorescence assays to investigate the subcellular location of the protein encoded by the DSCR2 gene in transfected cells. It was previously suggested that DSCR2 was located in the plasma membrane as an integral protein. Interestingly, we observed this protein in the endoplasmic reticulum of cells. We also studied whether the location of DSCR2 truncated forms had different subcellular distribution and our observations indicate that it is probably not inserted in inner membranes once the fragments lacking the predicted transmembrane helices remained in the ER. Se trata de un análisis institucional de organizaciones que tienen como fin evangelizar empresarios y profesionales liberales. El análisis concentrase sobre el producto que tais organizaciones ofrecen y también sobre los miembros de esas instituciones. Buscamos diseñar el perfil de tais organizaciones desde la caracterización de sus fundadores y de las historias de esas instituciones y, así descubrir eventuales cambios que habrían posibilitado la supervivencia y/o manutención de la misma. Tal análisis permitió identificar los objetivos de las organizaciones, su comportamiento dentro del campo religioso, la relación de ellas unas con otras, y con otros segmentos religiosos que representan de algún modo una especie de concurrencia en el mercado religioso.
