    Italianos em casos de conflitos e tensões nas fazendas de café da comarca de Araraquara, 1890-1914

    This study it approached cases of conflicts characterized for physical, moral, economic and racial the violence, for work questions, that involve deriving Italian immigrants of diverse regions of Italy and other ethnic groups on coffee plantations of the judicial district of Araraquara in the period of 1890-1914, using as source criminal records. The present work searched to understand the values that guided a violent behavior in the daily conflicts, investigating the solidarity possibilities between the ethnic groups in the individual actions, mainly between the Italians. In this perspective, the work evidenced that the regional differences between the Italians limited the solidarity of the group, over all of the northern ones and of central Italy in relation to the meridionals. The analysis of the criminal records also evidenced that the main values that guided a violent behavior in the daily conflicts were to receive a wage just and to have movement freedom. These values had been verified for all the ethnic groups in question.

    A Trajetória de uma escola cooperada: do ideal cooperativista à realidade de mercado. As vicissitudes da participação de cooperados e professores

    This work is case study researches about the trajectory of a cooperative school of basic education, located in São José do Rio Preto, inside of São Paulo state. The study has raised possible reasons in order to explain why in a school of cooperativist proposal and organization and in which parents and teachers who are supposed to take part in this process don t act as expected. Theoretical references have been studied in the national and international literature about cooperativism, political- pedagogical proposal, school administration and market politics. The dissertation begins with a chapter about cooperativist ideology. It goes over the cooperative and the educacional center, political-pedagogic process. The third chapter analyses the statutary alterations, the exhaustion of the assemblies in the last chapter we measure the capitalism and middle class market interferencies and implications in cooperatives in general, especially in COOPEC school. It is concluded that the cooperative democratic administration is seen as threatened by the lack of cooperativist ideology in parents and teachers, the directory staff authoritarism, the market lack of confidence and the reorganization of public school network.

    Espaços divididos e silenciados: um estudo sobre as relações sociais entre nativos e os "de fora" de uma cidade do interior paulista

    This study aims at understanding, through the methodological perspective of Oral history and making use of quantitative data, the construction and reproduction of multiple identities and sociabilities present among the rural workers natural from the Minas Gerais state and the Brazilian northeast region, and who have migrated to Guariba, a bedroom community in the interior of the São Paulo state whose economy is based on the sugarcane industry. In order to make this understanding possible, it has been necessary to determine in which contexts these identities and sociabilities are built, that is, how the surrounding community, with its ideas, thoughts and values, interferes in these social relations. It has been made clear to us that, between the two groups, there is a dichotomic and dialectic relation based on prejudice and symbolic violence, as well as a stereotyped connection between immigrants-criminality, which is clearer in Guariba due to a rural workers strike in 1984. This dichotomic relation is made possible by an ideology which permeates all the social groups in Guariba, and which divides the city between those we have called the natives (European descendants, downtown dwellers, middle class and white) and the outsiders (migrants from Minas Gerais and the northeastern states of Brazil, living in the suburbs, low class and black). When facing the stigma of the native group, the outsiders , being a heterogeneous group, have different and multi-faceted reactions, which can be subdivided in three groups: the seasonal migrant outsiders , the Guariba long-time living outsiders and the outsiders who belong to the second and third generations of migrants. Men and women who migrate yearly, during the sugarcane harvest, play different social roles in the cities where they are migrate to and their hometowns. When coming back to their home land, if they have been successful in the sugarcane plantations of the São Paulo interior, they are entitled to social and cultural differentiation according to their new identities and wealth. On the other hand, in the modern world where they have migrated to the relation is the opposite. The means of sociability are scarce and tense, with discriminatory basis, since for the native community the migrants are representatives of a traditional and undesirable world. The migrants that have lived in the city for more time are able to widen their sociability bonds, which spread in the suburban neighborhoods where they live. Nevertheless, they know there are places in the city where they are not welcome, and at the same time they recollect and revive their home land, where they feel really at home : the more often they migrate, the longer they stay in the same place. The representatives of the second and third generations of migrants, on the other hand, consider themselves (and in fact are) Guariba citizens, fruits of the modern relations in the São Paulo state, and for this reason they believe that all the places in the city belong to them by rights. However, because they are also considered outsiders , they easily notice the stigmas they are submitted to, and end up being more susceptible to violence relations, which makes us think that the natives symbolic violence is turned into real violence among this group. Finally, the migrants not only experiment the reunion of the different, but they live, above all, the inequalities and social differences of the cities and towns of the São Paulo state. The relation the migrants have with the surrounding community disguises color/race as well as social class prejudice against this group, which will never be us ; they will always be outsiders .

    Avaliação de fontes de carbono e condições de indução na expressão de canacistatina em Escherichia coli BL21 (DE3)

    Canecystatin (CC) is a competitive and reversible protease inhibitor which blocks the proteolytic enzymes activities of insets, fungus e nematodes, prejudicing thus the growth, development and reproduction from these pathogenics organisms. CC is produced by sugarcane, but with limited production, difficulting extraction and purification for production of commercials products, so recombinant DNA technology is one alternative for increase of CC production. Nowadays, CC is produced in the Molecular Biology Laboratory of the Department of Genetic and Evolution of UFSCar (LBM-DGE/UFSCar) from E. coli BL21(DE3) in shaker, utilizing a high cost commercial culture medium (Circlegrow®). With the purpose of produce CC in higher scale, the aim of present work was the study the CC expression in E. coli BL21(DE3), evaluating different carbon source, kind of inductor and induction moment in shaker, as well as confirming the expression conditions in airlift bioreactor of 6 L working volume. In the experiments carried out in shaker were obtained similar cellular growth when utilized glycerol, glucose, fructose e fructose + glucose, and low cellular growth when utilized galactose. It was observed high expression when galactose was utilized as carbon source and IPTG 0,4 mM as inductor. When lactose at 4 e 40 mM was utilized as inductor, CC expression occurred only in the culture containing galactose as carbon source. Volumetric production (in mg/L) of CC up to 61% those from standard culture was obtained, with exception the culture that utilized galactose induced with 0,4 mM of IPTG and 4 mM of lactose, and specific production (mgCC/gcells) greater 75% from standard culture. With respect of cultures in airlift bioreactor, CC expression was superior to expression culture carried out in shaker utilizing glucose with carbon source, approximately 80% of CC concentration obtained in the standard culture after 1 hour of induction and achieving more than 90% in the second hour of induction. In terms of specific production, in this culture was obtained 79,9 mgCC/gcell, value approximately 25% superior to the standard culture.

    Análise de metabólitos secundários em culturas de Pestalotiopsis sp e Aspergillus aculeatus

    Cariophylene sesquiterpenes are chemical compounds isolated from fungi of the Pestalotiopis genus in early studies developed in our research group. In the present work, a cultivation and analysis methodology were developed for these compounds in Pestalotiopsis, isolated from plants Pinus taeda, Swietenia macrophyla and Khaya invorenses. Different procedures were optimized for the extraction and analysis of the extracts employing liquid and gas chromatography coupled with mass spectrometry techniques (LC/MS and GC/MS). These studies had the intention of to detect the sesquiterpenes in fungal extracts without theys of isolation, and to check their production profile during a fixed cultivation period. The production of volatile organic compounds by the different species of Pestalotiospsis was also evaluated using hydrodestillation in Clevenger apparatus. The sesquiterpene cariophyllene was the main compound identified. Another study involving the secondary metabolites produced by Aspergillus aculeatus, a fungus isolated from the host plant Melia azedarach, was carried out. The target compounds in these studies were the dimeric aromatic poliketides Aurasperones A and B, Fonsesinones A and B, Orlandine and Kotanin. A poliketide production curve, obtained using HPLC, showed that the monomers are not detected during the early stage of development, indicating that probably the dimers are rapidly formed by the fungus or they are biosynthesized in the form already dimeric. Experiments of biotransformation showed that fungi possesss capacity to modify certain substracts, when cultivated in artificial ways.
