dc.description.abstractMolecular recognition at the interface between nanoparticles (NPs) and biomolecules depends on the structural and stereochemical organization of the interacting surfaces. However, the molecular mechanisms by which surface chirality modulates protein interactions remain only partially understood. This study investigated the influence of the chirality of trimanganese tetroxide nanoparticles functionalized with mannonic acid (Mn3O4-man) on their interaction with human serum albumin (HSA). Molecular dynamics simulations were performed to investigate systems containing apo-HSA and NPs with L- and D-chiral configurations. The trajectories were analyzed in terms of interaction energies, hydrogen bonds, intermolecular contacts, and structural and conformational parameters of the protein. The results showed that the interaction with HSA strongly depends on the chiral configuration of the nanoparticle surface, as only the L-NPs were able to establish stable and energetically favorable interactions with the protein. The D-NPs exhibited only transient contacts and interaction energy profiles close to zero, without the formation of persistent bound states. Spatial analysis of the interactions revealed that chiral recognition occurred locally, with Domain III of HSA, particularly the region corresponding to the Sudlow II site, being the only region capable of sustaining a stable interface with the L-NPs, mainly involving residues Lys538, Ala539, Lys541, and Glu542. The interaction energies and contact maps suggest that interface stabilization depends on the formation of a cooperative network of spatially complementary interactions, consistent with the concept of an interfacial “chiral footprint.” Conformational analyses indicated that persistent interaction with the L-NP induces localized dynamic changes, particularly in Domain III, without compromising the overall structural stability of HSA. The RMSD, RMSF, and radius of gyration results are consistent with a two-step recognition mechanism involving initial recognition driven by stereochemical compatibility between the surfaces, followed by local conformational adaptations consistent with an induced-fit mechanism. Overall, the results demonstrate that surface chirality acts as a selective structural factor in biomolecular recognition, influencing the affinity, temporal persistence, and spatial organization of protein-nanoparticle interactions. These findings contribute to the mechanistic understanding of nano-biomolecular interactions and provide a basis for the rational design of nanomaterials with enhanced biomolecular selectivity.eng
dc.description.resumoO reconhecimento molecular na interface entre nanopartículas (NPs) e biomoléculas depende da organização estrutural e estereoquímica das superfícies envolvidas. Entretanto, os mecanismos moleculares pelos quais a quiralidade superficial modula a interação com proteínas ainda permanecem parcialmente compreendidos. Este trabalho investigou a influência da quiralidade de nanopartículas de tetróxido de trimanganês funcionalizadas com ácido manônico (Mn3O4-man) sobre sua interação com a albumina sérica humana (HSA). Foram realizadas simulações de dinâmica molecular para investigar os sistemas contendo a apo-HSA e NPs nas configurações quirais L e D. As trajetórias foram analisadas quanto às energias de interação, ligações de hidrogênio, contatos intermoleculares e parâmetros estruturais e conformacionais da proteína. Os resultados mostraram que a interação com a HSA depende fortemente da configuração quiral da superfície nanoparticulada, uma vez que apenas as L-NPs foram capazes de estabelecer interações estáveis e energeticamente favoráveis com a proteína. As D-NPs apresentaram apenas contatos transitórios e perfis energéticos próximos de zero, sem formação de estados ligados persistentes. A análise espacial das interações revelou que o reconhecimento quiral ocorreu de maneira localizada, sendo o Domínio III da HSA, particularmente a região correspondente ao sítio Sudlow II, o único capaz de sustentar uma interface estável com as L-NPs, envolvendo principalmente os resíduos Lys538, Ala539, Lys541 e Glu542. Os resultados energéticos e os mapas de contato sugerem que a estabilização da interface depende da formação de uma rede cooperativa de interações espacialmente complementares, compatível com o conceito de “pegada quiral” interfacial. As análises conformacionais indicaram que a interação persistente da L-NP promove alterações dinâmicas localizadas, especialmente no Domínio III, sem comprometimento da estabilidade estrutural global da HSA. Os resultados de RMSD, RMSF e raio de giro são compatíveis com um mecanismo de reconhecimento em duas etapas complementares, envolvendo um reconhecimento inicial dependente da compatibilidade estereoquímica entre as superfícies, seguido por adaptações conformacionais locais consistentes com mecanismos do tipo encaixe induzido. Os resultados obtidos demonstram que a quiralidade superficial atua como um fator estrutural seletivo no reconhecimento biomolecular, influenciando a afinidade, a persistência temporal e a organização espacial das interações proteína-nanopartícula. Esses achados contribuem para a compreensão mecanística das interações nano-biomoleculares e fornecem fundamentos para o design racional de nanomateriais com maior seletividade biomolecular.por
