This work describes the development of electrochemical immunoassays using electrode disposable devices made from low cost materials as an alternative for detection of the biomarkers p16, GSTP1, PSA and CYFRA-21-1 for prostate cancer diagnosis. In the immunoassays developed, magnetic micro/nanoparticles were used in order to provide the capture and separation of the biomarker and the amplification of the analytical signal allowing to reach high selectivity and low limits of detection. For this propose, disposable microfluidic cell was developed and constructed containing 8 carbon-based working electrodes, an Ag|AgCl electrode as a pseudo reference electrode and a carbon electrode as an auxiliary electrode. Three different immunoassays were developed. The first work consisted in the development of an electrochemical immunoassay for the detection of the p16 biomarker. Immunoassays were performed after optimization of the parameters, such as incubation time and flow rate used in the microfluidic system. The calibration curve was prepared in calf serum with p16 concentration in the range of 1.38 to 27.7 fg mL-1. The device presented detection limit (LoD) of 0.04 fgmL-1 and sensitivity of 11.6 nA (fg/mL)-1. The second method consisted of the simultaneous detection of PSA and GSTP1 biomarkers. Peak currents obtained showed a linear response between 0.7 - 27.8 fg mL-1 for GSTP1 and 0.28-277 fg mL-1 for PSA. The sensitivities reached for GSTP1 and PSA were 22.4 nA (fg/mL)-1 for both biomarkers and the LoDs were of 0.15 fg mL-1 for GSTP1 and 0.16 fg mL-1 for the PSA. Finally, the third immunoassay consisted of the application of synthetic nanoparticles (MNPs) of Co0.25Zn0.75Fe2O4 (CoZnFeONPs) and Fe3O4 (FeONPs) used to mimic the enzyme peroxidase. The use of modified carbon work with graphene oxide (GO) and MNPs allowed the electrochemical detection of CYFRA-21-1 with LoDs of 0.30 fg mL-1 and 0.19 fg mL-1 for FeONPs and CoZnFeONPs, respectively. Finally, it was possible to test the immunosensor in serum of patients provided by the Hospital de Amor from Barretos city obtaining concordant results with ELISA method, which showed that the device is a promising choice for diagnosis of prostate cancer in relation to traditional methods as ECL and ELISA.

The water crisis is a hotly debated issue in recent years, especially as regards the effects of globalization, both on the interference of climate change directly on recharge and on the disorderly increase of the urban environment that directly affects the availability of water for domestic consumption, and industrial. These climate changes alter pluviometric regimes and volumes, evapotranspiration, the recharge process of aquifers, and river flows. Deforestation and paving of recharge areas are the result of urbanization and the advancement of agricultural production, leading to increased waterproofing of the soil and thus affecting the infiltration and storage of groundwater. The objective was to fill some gaps in the literature, in relation to the studies that investigate the water refills, using a bibliometric mapping and geological-geotechnical detail mapping, aiming the monitoring of water recharge zones in the region north of the municipality of São Carlos, SP. In order to do so, the monitoring of free aquifer of the microbasin was carried out by means of frequent measurements of the static levels of 4 monitoring wells for the subsequent application of the Water Tabel Fluctuation (WTF) method that verified an annual recharge of 1338.8mm, lower value to the previous years, representing an average of 16-36% of all direct annual recharge of the microbasin. In addition, a multitemporal analysis of the soil cover type was done in this study, demonstrating the problems generated by the increase of the percentage of soil exposed in regions of springs, increasing, consequently, the surface runoff and reducing the infiltration. It is concluded that the monitoring of the recharge of water has been satisfactory and that stricter measures in the management of water resources must be taken so that future generations do not suffer from water crises in the region. In addition, urban sprawl in the study area may directly interfere with this decrease in surface recharge.

The understanding of autism, currently known as Autism Spectrum Disorder (ASD), has undergone several changes over the years. In the Diagnostic and Statistical Manual of Mental Disorders (DSM-III), in 1980, came the inclusion of autism as Pervasive Developmental Disorders (PDD). However, in the tenth revision from the International Classification of Diseases (ICD-10) from the World Health Organization (1992), it included childhood autism inside the global developmental disorders group. At the DSM-IV (1994), in addition to autism, was the inclusion of the Asperger syndrome and Pervasive developmental disorder not otherwise specified, and, in the fifth and latest version (DSM-V), from 2013, the denomination used was "autism spectrum disorder"; besides changing the denomination, occurred the modification in the structure of symptoms and signs group that determinates the diagnosis of ASD. In Brazil, philanthropic institutions within social services or autistic family associations are often responsible for the care of these children with autism. Some studies show that this late development caused the elaboration of the public policy for autism to be marked by two opposing groups: the group linked to the actions from the psychosocial attention related to the public policy of the unified health system (SUS), and the group associations of parents and family members of autistic people. With SUS public policies, were created the Child and Youth Psychosocial Care Centers (CAPSij) with the perspective of territorial health and articulation with the Psychosocial Care Network (RAPS). Recent studies address the difficulties encountered in these CAPSij concerning strategies and actions aimed at these children and adolescents with autism, highlighting the need for research to assist in the identification and understanding of the demands that reach these services. The current study aimed to characterize people with autism-linked to a CAPSij in a town in the state of São Paulo. It is an exploratory research of a quantitative approach that involved the participation of managers from all four Child and Youth Psychosocial Care Centers (CAPSij) from a town in the state of São Paulo. For data collection, modules of a structured interview script were used, elaborated in the broader research to which this TCC is linked, and a medical record data record form, elaborated in this study. It revealed that 26% of the CAPSij patients were autists, with a prevalence of reach between 1 and 5 years old, mostly males. The study also identified some weaknesses in the engagement of adolescents with ASD in these services. In general, the results achieved revealed fragilities resulting from the team composition in CAPSij, that should impact the care practices and actions related to ASD in particular.

(Universidade Federal de São Carlos, 2019-04-05) Lopes, Natalia Roberta; Godinho Filho, Moacir; https://lattes.cnpq.br/6136685270563354; https://lattes.cnpq.br/3575543749691837
Lean manufacturing continues to be widespread in the business world as a management philosophy to reduce waste. Despite this, the sustainability of the results of lean initiatives has proved difficult to achieve. There is a limitation on the part of organizations in understanding the factors that influence lean sustainability in all organizational environments. In this direction, the objective of the research was to identify the critical factors for manufacturing companies to sustain their Lean initiatives over time. For this the mixed research method was used, combining the systematic review of the literature, case study and expert opinions. In the systematic review, articles were filtered in scientific journals: Scopus, Engineering Village, Proquest and Web of Science in order to identify in the literature the factors mentioned as critical to sustain lean. Thus, combining the review with snowball methods and expert opinion, we obtained a list of 21 factors. This list was analyzed in the case study with two industries for refinement of the list. Finally, the list of factors passed by the analysis of 5 experts, in the end 19 factors were considered as critical. The results achieved contribute to the professionals have a vision of what aspects should be addressed throughout the Lean journey. In the academic context, the contribution was to compile the lean sustainability factors that are spread in the literature and to show which ones should be explored.

Ferroelectrics are material exhibiting a spontaneous polarization that may be switched by an electric external field. This behavior is provided to well-known ferroelectric domain walls motion. Recent research has proved the surprising ability to move ferroelectric domain wall under illumination with wavelength of 532 nm incident on a surface of single crystals of Barium Titanate (BaTiO3). In spite of experimental results concerning about this light-matter interaction, it is remarkable a lack of physical insight, as well as possibility to observe if occurrences of motion domain wall is due to single or poly crystal character. By using a phenomenological approaches based on Devonshire-Landau’s theory was possible make a development to describe a polarization-induced switching under light like perturbation of bound and free charge accumulated within domain wall. Accordingly, interactions between photons having lower energy that optical gap and screening charge give rise to fluctuation in depolarization electric field that serve as a driving force to polarization reorientation around the wall. This approach gives a satisfactory explanation to experimental facts observed in single crystals of BaTiO3, in addition, it allows to conjecture a correlation between the phenomenon occurrence with existence of charged domain wall as single crystal as polycrystal. It was prove that BaTiO3 coarse grain ceramics (grain size approx 50µm) contend 90-domains are possible a manifestation of polarization switching induced by light also, in this form, it was demonstrate the size grain dependence with evidence of this light-matter interaction, highlighted again the role-play of charged ferroelectric domain wall. In this way, here has made an extend to polycrystal materials that could lead to developed the next generation of photo-stimulated ferroelectric devices.
