Two process variables, namely the glucose and inoculum concentrations, were evaluated in different levels (5 to 40 g/l, and 2.5% to 5.0% v/v, respectively), and their effects on biomass formation, minimal inhibitory concentration (MIC) against Escherichia coli and surface tension reduction (STR) were studied. The fermentation process was firstly carried out using non-optimized parameters, where the dependent variables biomass, MIC and STR reached the values of 0.6 g/l, 1.000,0 μg/ml and 18.4 mN/m, respectively. The optimum glucose (16 g/l) and inoculum concentrations (5.0% v/v) were defined in order to maximize the biomass formation, with low value of MIC and large STR of extract. Under these conditions, a biomass of 2.76 g/l, MIC of 15.8 μg/ml, and STR of 14.58 mN/m were predicted by the model. Data attained by experiments using optimized settings showed the following values: biomass 2.05 g/l; MIC 31.2 μg/ml; STR 10.7 mN/m. Thus, the percentage of improvement for each target response was: biomass 241.6%; MIC 96.88%; STR 41.85%. It was found that high concentrations of glucose substrate, although reflected in an increase in bacterial biomass, inhibited the microbial secondary metabolism, resulting in a low production of biomolecules associated with high values of MICs. Thus, initial concentrations of glucose and inoculum are shown as variables of strong influence in the production of antimicrobial metabolites by P. polymyxa RNC-D. Through the methods of experimental factorial design and surfaceresponse followed by graphical optimization it was possible to determine the optimum operating condition to achieve both maximum biomass and RTS as well as and lowest possible values of CIM. The validity of the proposed model was verified and confirmed. This is the first study on the optimization of culture conditions for the production of antimicrobial metabolites by P. polymyxa RNC-D, and constitutes an important step in the development of strategies to modulate the production of antimicrobial molecules by this microorganism in elevated levels. Novel antimicrobial compounds were isolated from the fermentation broth of P. polymyxa RNC-D, here named total extract (TE). It was possible to verify the presence of lipopeptide and peptide active compounds through enzymatic assays made with ET. Total extract was subjected to a two-phase system, resulting in lipopeptide extract (LPE) and aqueous fraction (AF). According to the results of bioassays, LPE has broad-spectrum activity against Gram-positive bacteria, Gram-negative bacteria and fungi. The mass spectrometry analysis of PLA revealed the existence of a novel compound that was named polycerradin. The purification of a novel antimicrobial peptide (AMP) from the AF was carried out by using chromatography. The compound was active against Gram-negative bacteria. Nterminal analysis determined the amino acid sequence, as well as MS / MS analysis confirmed the primary structure of this new compound. This research reports firstly the production of PAM PpRNCD that has an unusual amino acid in its constitution. It is an unprecedented fact considering the bacterial specie P. polymyxa. In terms of molecule size, PAM PpRNCD can be considered one of the smallest active natural peptide reported to date. It was also possible to isolate from FA the depsipeptides IL-F04a (m/z 883), LI-F04b (m/z 897), LI-F03a (m/z 947) and LI-F03b (m/z 961) previously described in the literature. The photoluminescence study of the LPE, TE, AF in both at room temperature (RT) and low temperature (T = 8K) was performed. In addition, this technique was applied to evaluate the action of the ELP on Staphylococcus aureus ATCC 29213, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 29212, Shigella sonnei ATCC 1578 and Candida albicans ATCC 10231 in two different situations: (a) immediately after mixing LPE with the bacterial and fungus cell suspension, and (b) after thirty minutes. The photoluminescence emission was collected by a triple spectrometer (three diffraction gratings) T64000 model from Jobin Yvon, equipped with an optical microscope. For the detection of the radiation emitted by the sample we used a CCD camera (charge coupled device) cooled by liquid nitrogen. The slits of the spectrometer were adjusted to produce a spectral resolution of the order of 10-4 nm. The excitation source used was the line of 457 nm (violet) from an argon laser. Like other bimodal respirators, the African catfish presents cardiorespiratory adjustments to survive low PwO2. These regulations are modulated by gill O2 chemoreceptors monitoring the PwO2 and/or the PaO2. The present study analyzed the distribution, location and orientation of the branchial O2 chemoreceptors of C. gariepinus, and the role of these receptors on the cardiorespiratory function and the modulation of the heart autonomic control. C. gariepinus were divided into two experimental groups (Control GC, intact fish, and Experimental GD, with the 1st gill arches excised) and their cardiorespiratory variables were analyzed during graded hypoxia with and without access to the atmospheric air, internal and external NaCN injections and autonomic nervous system antagonists. During hypoxia without access to air, both GC and GD showed increases in the respiratory variables (fR, VT and 𝑉 𝐺 ). However, the GD displayed higher values, since normóxia until severe hypoxia, to compensate the decrease in the EO2 resulting from the ablation of the 1st gill arches. Furthermore, the decrease in EO2 was responsible for the decrease in 𝑉 O2 and, consequently, for the increase of the PcO2 from 61 mmHg (GC) to 79 mmHg (GD). The GD featured bradycardia similar to GC, however, their values were higher in all PwO2. In hypoxia with access to air, both groups showed decrease in the fR and VAMP in the mores hypoxic tensions. This was attributed to the increases in the fRA in GC which compensate the O2 uptake. However, the GC increases in fRA were attenuated by the excision of the 1st gill arches. Moreover, both groups developed bradycardia pre-RA (typical of aquatic respirator) and tachycardia post-RA (typical of aerial respirator). Internal and external NaCN injections caused bradycardia in both groups, while the internal injections increased the ventilator variables only in GC. Thus, it is concluded that C. gariepinus possess branchial O2 chemoreceptors modulating the fH distributed by all gill arches, monitoring both PwO2 and PaO2. Conversely, fR and VAMP are modulated by receptors located in the 1st gill arches monitoring exclusively the PaO2. Furthermore, NaCN internal injections elevated more the fRA in GC than the external ones, but with low effect on the fRA of GD. Thus, the fRA is primarily modulated by O2 chemoreceptors located in the 1st gill arches monitoring, predominantly, the PaO2. Injections blocking the autonomic system to the heart showed that the modulation of pre-AR bradycardia and post-AR tachycardia occurs due to variations in the vagal cholinergic tonus, which is elevated during bradycardia and reduced during tachycardia. The adrenergic tonus, in spite of the decrease in the post-AR, is lower than the cholinergic, suggests a "reflex adrenergic" arising from the peripheral vasoconstriction, important to optimize the perfusion of the ABOs.listelement.badge.dso-typeItem, Identificação de regiões genômicas e genes candidatos posicionais e funcionais para características de eficiência alimentar em bovinos da raça Nelore(Universidade Federal de São Carlos, 2014-06-30) Oliveira, Priscila Silva Neubern de; Regitano, Luciana Correia de Almeida; https://lattes.cnpq.br/2168962905598747This thesis has been divided into three chapters for better understanding of the experiments. The first chapter refers to a brief Literature Review, with the main concepts and justifications for this work. The second chapter discusses the strategy of positional and functional candidate genes for traits of feed efficiency in Nelore cattle, and the third chapter presents a genome-wide association study for feed efficiency traits in the same population. Feed efficiency is an important production trait in beef cattle production systems. The aim of this study was to identify genes/QTLs associated with feed efficiency traits in Nelore cattle genotypes using the Illumina BeadChip BovineHD (770K) of 593 Nelore steers. The traits analyzed were: average daily weight gain (ADG), dry matter intake (DMI), feed conversion (FC), feed efficiency (FE), residual feed intake (RFI), efficiency of maintenance (EM), efficiency gain (EG), partial efficiency of growth (PEG) and relative growth rate (RGR). The first strategy discussed in this work, using candidate genes, investigated the association of Neurogenic differentiaton 1 (NEUROD1), Kv channel interacting protein 4 (KCNIP4), and Vascular endothelial growth factor C (VEGFC) genes, with production traits in the same population. Our results are in according to literature that indicate NEUROD1, KCNIP4 and VEGFC as candidate genes for feed efficiency; and this study is the first to suggest the NEUROD1 gene as a candidate for gene backfat thickness, body weigth and metabolic weight in Nelore cattle. In the genome-wide association study some QTL regions identified in this study minimally overlapped with QTL regions previously reported for feed efficiency in Bos Taurus cattle, and harbor genes with biological functions related to metabolic processes, lipid and protein metabolism, energy generation and growth. A comparison with published results indicates that different QTL and genes may be involved in the control of feed efficiency in this population of Nelore cattle. Marked by an ambivalence of vocations, the Brazilian Bar Association managed to move self confidently between corporate and institutional interests. It is by taking advantage of this characteristic and its history, mainly related to conflicts of legal and democratic nature, that we are motivated to immerse into a universe of this sui generis institution, drawing Law and Policy closer in such a way that they seem inseparable. The chosen arena, the Constituent of 1987, brings all the characteristic features of a good political novel , full of alliances and strategies. The topic that we report as ideal is the analysis of questions which involve setting up a Constitutional Court, proposed by the association, which went through procedures in the Subcommittee of the Judicial Power and the Public Prosecutor. One of the main obstacles was the centuries-old tradition of the Supreme Constitutional Court, an organ of the Judicial Power, which would be substituted, if the progressive intent of the Bar Association in Brazil had been successful. Moreover, taking this into account, we attempt to clarify which factors contributed to this frustration, even knowing that approximately 75% (seventy five percent) of the constituents belonging to the Subcommittee were professionally declared as lawyers.listelement.badge.dso-typeItem, Ação ergonômica e estratégias de operações: proposta de integração na prática(Universidade Federal de São Carlos, 2014-06-30) Gonçalves, Juliana Machion; Camarotto, João Alberto; https://lattes.cnpq.br/1427853773681255; https://lattes.cnpq.br/4059272741028061This research has, with main objective, the construction of activity ergonomics management model that is related operations strategies, developed from practical experience in the company with metal - mechanics production of white goods, involving a group of approximately 1.500 people and 400 workplaces, aimed at structuring an ergonomic action, based on the activity ergonomics, which it is aligned with the operations strategy and understanding the consequences (positive and negative) about ergonomics action, the perception of the operators and managers of the company, the relationship between health and work. The construction involved a participatory process the transformation of the working conditions and workplace conception. The theoretical-methodological approach has the assumptions of ergonomics work analysis and action research were used to construct the model based on the activity ergonomics of operations strategies related in order to emerge among all the actors involved greater understanding of the work, its determinants and constraints, as well as the instrument to represent the perceptions of the actors of this process and its contradictions. The demand started from high rates of removal by musculoskeletal disorders in the area of manufacturing company. The data were indicators that guided the certificates and clearances. Training, analysis / diagnosis and projects were the steps of action research. Relation about training, a ergonomics committee was formed with the performance of health professionals, process engineers and production supervisors. These actors were trained and helped to survey the descriptions of the task, the constraints involved, the rules and expected results and project items, so they would get to the point of view of activity and operations strategies, to further suggest recommendations necessary. Moreover, the process of analysis / diagnosis was developed from the ergonomics law (NR 17) with assumptions of ergonomic work analysis (AET). Finally, the stage was set designs from the results of a summary of the analysis / diagnosis. Every problem encountered was given a solution, aligning with operations strategies (organization of work, performance measures, process technologies, and human resources) through implementation of breaks, job rotation, procedure for development projects, order to modify the organization of work and technical and organizational design and in case devices, design of design processes and correction operations. The manufacturing management must seek the proper standardization of operations, compliance with these standards and continuous improvement, using this as a basis for synchronization of material flows and information, and to guarantee the quality and safety requirements, as well as the increase responsibilities and autonomy of people in the workplace. This is the main base of lean manufacturing (lean), which, associated with the concept of semi autonomous teams (this aimed at increasing the responsibility of the workers), significant operational success is bringing to companies that have deepened in its implementation and support. This thesis presents six case studies where there was the application of a model of standardized work and the concept of semi-autonomous teams simultaneously in different plants of the same company in the Brazilian chemical industry. The developed approach was the adaptation and application of a systematic model with five pre-defined steps, and structuring teams with semi autonomous work patterns as well as their roles and responsibilities. The model applied in the six cases studied was adapted based on a reference from the company's headquarters and research of market practices, adapted to reality and to the organizational structure of these units. The plants studied have different levels of technology and products portfolio, as well as operation time that distanced up to four decades of each other, where it was observed significant impact on speed of deployment, with different levels of acceptance by the leadership and comprehensiveness of results arising from the application of the model. Quantitative aspects of implementation, such as Safety, Quality and Productivity, which showed significant weight gains after the implementation of the model were evaluated and achieved medium gains of 25% in Productivity, 33% in Quality and 47% reduction in setup times as well as qualitative aspects such as absenteeism, the perception of those involved on the shop floor by the physical change in the working environment and the impact of standardization and systematization of activities on the motivation and performance of employees working in the semi autonomous teams. The participants are pre-service teachers of English as a foreign language, who are in the third-year of their undergraduate course in Languages and Literature. The theoretical frameworks of this research combine the theory and pedagogy of genres and the theory of teacher education, the latter in light of the teacher cognition studies. The investigation can be classified as essentially qualitative, with an interpretive and ethnographic perspective. The data analysis process reveals that the use of the genre theory in language teacher education can be considered an adequate instrument for both, a better understanding of the text, ultimately promoting a better understanding of the situated use of the target language by student-teachers, as well as an exercise of methodological choices and implementation of education. These aspects, together, converged on an improvement of teacher knowledge. For this, executest were three main steps: 1) The choice of the species of study; 2) Review the unit of study with different models for the species of study; and 3) Propositions for the unit of study. The choice of the species of study was based on a questionnaire to various ornithologists, where we tried to select the most suitable species for the study. For the analyzes of the study unit was used , on the conception of Landscape Ecology , tools such as scanning a satellite image obtained by Google Earth ® and landscape analysis by MapInfow ® program. Propositions for the unit of study was based on analysis of the model considered more real and critical to the species of study aiming at establishing minimum viable conditions for the establishment and maintenance of the species studied. This specie of study was chosen Antilophia galeata, the unit of study is located in the municipality of São Carlos (SP), and the whole basin of Ribeirao das Araras. This study showed that one must have a very great care in analyzing the most appropriate model for a given species, because the way that the landscape can be analyzed can generate completely different results and sometimes lagged the actual situation, as evidenced by this paper.
