    Objectives: The main aim of this thesis was to investigate whether personalized circuit training promotes effects on pain processing, determined by pressure pain thresholds, temporal summation, and conditioned pain modulation in patients with knee osteoarthritis (KOA), compared to an educational control group. Secondarily, the objective of this study was to investigate whether there is a correlation between quantitative sensory measures and sleep quality, as well as with sleep components, and to evaluate the feasibility and acceptability of this protocol. Format: For this purpose, three studies were conducted. The first one explored the relationship between sleep quality and its components and pain sensitization and was entitled "Pressure pain threshold and temporal summation predict poor sleep quality in knee osteoarthritis". The second one analyzed the feasibility and acceptability of the protocol and was titled "Tailored Circuit Training and an educational program reduce pain sensitivity in patients with knee osteoarthritis: a feasibility study". The third study presents the results of the clinical trial, titled "Remotely-delivered personalized circuit training reduces pain sensitivity in patients with knee osteoarthritis: A randomized controlled clinical trial. "Methods: For all three studies, participants with a primary symptom of pain (≥ 4) and KOA diagnostic criteria were recruited. They were randomly allocated to an experimental group or a control group in a 1:1 ratio for 14 weeks of intervention delivered remotely and a total of 26 weeks of follow-up. The experimental group performed a personalized circuit training program and an educational program, performing exercises three times a week, with individualized progression of exercise duration and intensity. The control group performed only the educational program, which included seven meetings, held every two weeks, in which they received information on the pathophysiology of KOA, treatment recommendations, and topics in pain neuroscience. At baseline, after the 14-week intervention and after the end of follow-up, at week 26, clinical measures such as physical performance, self-administered questionnaires on physical and mental condition, and muscle strength, and quantitative sensory tests were collected. For study 1, only baseline data were used, since it is a cross-sectional study. Open and closed questions were asked for study 2, about feasibility and acceptability. Studies 1 and 2 were analyzed with descriptive statistics, correlation tests and linear regression analysis, with the SPSS 26.0 program and a significance level of 5%. Statistical analyses of the clinical trial were performed using the R programming language. The between-group differences and 95% confidence intervals for post-treatment outcomes were calculated using the linear mixed models through the groups-versus-time interaction analysis. Results: The first study included 80 participants and identified that temporal summation, local pressure pain threshold, and depression and anxiety were associated with poor sleep quality, specifically the components of subjective sleep quality, sleep duration, sleep efficiency, sleep disturbances, and daytime dysfunction. The second study included 30 participants and identified an eligibility rate of 31.6% and a recruitment rate of 2.7 participants per week, considered adequate. Adherence to the exercise protocol was partially adequate (60%), and the educational program was adequate (80%). Differences between groups at different time-points for primary and secondary outcomes were presented. Finally, the clinical trial included 88 participants, 60% of whom were female, and the average age was 59 years. The mean difference between groups in pressure pain threshold in the medial line of the knee was increased (1.3 kg/cm2 [95% CI from 0.8 to 1.7]) in relation to the control group, as well as the other pressure pain thresholds and their CIs. The estimated differences in temporal summation and conditioned pain modulation had reduced means, but wide confidence intervals, and the results were considered inconclusive. Among the clinical parameters, a reduction in pain intensity (-2.3 [95%CI -3.3 to -1.4]) and symptoms of KOA by WOMAC (-14.4 [95% CI -19.8 to -9.0]), increased strength of the knee extensor muscles (4.8 Kgf [95% CI 2.7 to 6.8]) and improved physical performance for climbing and descending stairs (-4.9 s [95%CI -7.6 to -1.9]) were identified in the experimental group compared to the control group. An association between pain intensity and progression in the exercise protocol was identified. Conclusion: The protocol of assessments and interventions with remotely-delivered personalized circuit training and patient education achieved good rates of feasibility and acceptability. Anxiety and depression, and peripheral pain sensitization were associated with poor sleep quality, specifically sleep components, subjective quality, duration, sleep efficiency, sleep disturbances, and daytime dysfunction. Remotely-delivered personalized circuit training constitutes an effective strategy for modulating pain sensitivity and clinical symptoms in individuals with KOA. Higher pain intensity was determinant for slower progression in the exercise protocol.

    Ativação in situ de oxidantes mediada por irradiação UVC: um estudo do tipo e concentração de oxidantes em distintas matrizes aquosas

    As a consequence of the increasing production and use of synthetic organic compounds, a growing detection of these pollutants has been observed in water bodies. Depending on their concentration and exposure, these substances may pose risks to human health and aquatic life. One of the main factors contributing to their presence is the inefficiency of conventional treatment methods, which are not effective in removing such compounds. In this context, the present study investigated the efficiency of UVC-based advanced oxidation processes for the simultaneous removal of four micropollutants: ciprofloxacin (CIP), enrofloxacin (ENRO), norfloxacin (NOR), and imidacloprid (IMD). Two systems were evaluated: UVC/H2O2, as a source of hydroxyl radicals (HO•), and UVC/S2O82–, as a combined source of sulfate (SO4•–) and HO• radicals. For the three fluoroquinolones, a concentration of 500 μmol L–1 of H2O2 provided the best results in terms of oxidation rate and oxidant consumption. In the case of the UVC/S2O82– system, its performance was similar across all tested concentrations, allowing the same value to be used for comparison. Overall, the UVC/S2O82– process showed superior performance compared to UVC/H2O2 in the oxidation rate of fluoroquinolones. On the other hand, the oxidation of IMD was not significantly affected by the different oxidant concentrations, resulting in similar oxidation rates for both processes and being primarily dependent on the photochemical process. The efficiency of both systems was also evaluated in different aqueous matrices: deionized water, tap water, and simulated effluent from a biological wastewater treatment plant. In the UVC/H2O2 process, a slight inhibition in the degradation of micropollutants was observed in the more complex matrices, especially in the simulated wastewater, likely due to light attenuation and the consequent decrease in oxidant activation. In contrast, in the UVC/S2O82– system, both the oxidation rate and oxidant consumption remained similar across all matrices, demonstrating greater robustness and versatility in the presence of matrix interferences. Toxicity assays using Escherichia coli indicated that although UVC irradiation alone achieved a good oxidation rate, it was not effective in removing the residual toxicity of fluoroquinolones. In contrast, the UVC/H2O2 and UVC/S2O82– processes proved effective in reducing toxicity, reinforcing their potential for application in advanced treatment of effluents containing emerging contaminants. Finally, process cost was shown to be largely dependent on the lamp power and the type of pollutant targeted for oxidation.

    Air quality and reuse of plastic waste are two problems that have challenged humanity and the terrestrial ecosystem. Expanded polystyrene (PS-E) can be recycled to produce air filtering media to, simultaneously, address these two problems. Thus, this work had as objective to develop air filters from recycled PS-E produced via electrospinning. Process variables such as polymer concentration, spinning time, type of collecting substrate, and the addition or not of ionic surfactants were investigated. The influence of the collecting substrate on the formation of nanofibers was observed, the nylon mesh resulted in a reduced area of fiber deposition and incomplete evaporation of solvent. Samples spun on steel mesh maintained surface electric potential (TES) even after 30 days, while those electrospun on copper mesh have their surface voltage completely dissipated after 7 days. An atypical phenomenon was observed in some samples that presented momentary increase of TES, possibly related to the surface redistribution of charges. Electret filters presented overall efficiencies higher than 90% for particles with average diameter of 0.25 μm (MP0,25), with quality factors (QFs) varying between 0.035 and 0.064 Pa⁻¹. However, after electrostatic discharge, the overall efficiency was reduced to values between 52.26% and 76.89%, and the QF decreased to the range of 0.008 to 0.024 Pa⁻¹. The polarity of the voltage applied during electrospinning does not appear to have effect on the collection efficiency, but affects the morphology of the fibers, with the samples of negative polarity presenting greater occurrence of structural defects. The addition of ionic surfactants cetyltrimethylammonium bromide (CTAB) and sodium dodecyl sulfate (SDS) resulted in absence of TES (0 kV), indicating that the increase of the conductivity of the solution facilitated the escape of the charges. Despite the absence of the electret character, these filters exhibited high collection efficiency, higher than 99.2% for MP0,25. Moreover, the formation of self-assembling fibrous stacking as well as the presence of electrowebs was observed. These results contribute to the understanding of production of electret filters and reinforce the possibility of employment of recycled PS-E as raw material for the production of air filtering media.

    Dictyoloma vandellianum is a species of the Rutaceae family known for its diversity of secondary metabolites with biological potential. This study aimed to characterize compounds present in leaf extracts of the plant, focusing on the prospecting of substances with potential antibacterial activity against Xanthomonas citri subsp. citri, the causal agent of citrus canker. Crude extracts were obtained using solvents of different polarities and subsequently analyzed by HPLC-PDA to assess the chromatographic profile. A detailed analysis of the constituents was carried out by UHPLC-QTOF-MS/MS in both ionization modes, followed by data processing using metabolomics platforms (MS-DIAL, MS-FINDER, and GNPS), with in silico annotation and molecular networking. This approach revealed the presence of compounds belonging to the classes of flavonoids, alkaloids, quinolones, and coumarins. Three substances were isolated through chromatographic fractionation and structurally characterized by one- and two-dimensional NMR spectroscopy, with validation based on fragmentation patterns observed in mass spectrometry. Although the identified structures are not novel, they were confirmed with a high level of confidence using the employed analytical methods. The results reinforce the potential of D. vandellianum as a source of bioactive metabolites and provide a solid foundation for future investigations. Next steps include testing the isolated compounds against the phytopathogen and structural characterization of additional constituents yet to be identified.

    Vestindo sentidos: enunciados aderentes sobre parentalidade em camisetas

    Dans le marché contemporain, il y a une circulation croissance de déclarations d'adhésion sur des objets quotidiens tels que les T-shirts, dont le langage mobilise des significations sociales et identitaires. Dans cette étude, nous proposons d'analyser, sur un site web, comment les significations de la parentalité sont configurées dans les énoncés adhérents imprimés sur les Tshirts Digorestte, en nous appuyant sur l'Analyse du discours dans une perspective énonciative, et notamment sur les apports théoriques de Dominique Maingueneau (2013, 2015, 2020, 2022). Nous cherchons ainsi à comprendre comment les énoncés adhérents sur la parentalité actualisent des pratiques discursives qui traversent l'espace public et la consommation, et comment ils construisent un ethos qui interpelle les sujets-mères et les sujets-pères. L'analyse est guidée par les questions suivantes : comment les énoncés adhérents sont-ils planifiés à l'avance par le site de la marque pour interpeller un sujet encore générique ? Comment s'établit l'adhésion entre les dictons et le support du tee-shirt, surtout dans un moment qui précède l'appropriation individuelle - ce que nous proposons d'appeler la pré-adhésion ? Enfin, quels réseaux interdiscursifs traversent ces énoncés et soutiennent les formes de reconnaissance sociale de la parentalité ? Pour traiter ces réflexions, nous avons également mobilisé les contributions d'auteurs tels que Neyrand (2011), Zornig (2010), Butler (2004), Schneider (2011), Campos et Tílio (2020) et Bauman (2001), avec lesquels nous avons discuté des notions de parentalité, de performativité, de reconnaissance et d'idéologie. L'analyse montre que les énoncés des adhérents sur la parentalité agissent comme des dispositifs discursifs qui (re)produisent des normes sociales et des valeurs associées aux soins, à l'autorité et à la protection, mettant en tension la place des pères et des mères dans la société contemporaine.

    Cette recherche a pour objectif de mener un laboratoire de transcréation de Dons des Féminines (1951), de Valentine Penrose, œuvre dans laquelle la poétesse articule trois éléments essentiels : la langue française, la langue anglaise et le collage. Pour appuyer notre analyse, nous abordons l’acte traductif comme un espace privilégié de réflexion critique sur la poétique de Penrose, ainsi qu’un terrain d’expérimentation pour la transcréation de l’œuvre (Campos, 2005). La transcréation se conçoit ici à la fois comme méthode et comme cadre théorique. Il s’agit de vérifier chez Penrose l’existence d’une « écriture-collage » (Colvile, 2001), une écriture qui transgresse le langage littéraire en recourant à des procédés visuels. La démarche de création et d’analyse critique permettra de dégager les caractéristiques sémantiques, formelles et plastiques de l’œuvre, en vue d’analyser Dons des Féminines comme un corps unique, où les langages déployés produisent du sens dans leur articulation même.
